Objective:To investigate expression of FasL and Caspase-8and FADD in non-small cell lung cancer tissues, corresponding paracancerous tissues as well as in metastatic lymph nodes and non-metastatic lymph nodes. Analyze the correlation of FasL, Caspase-8and FADD expression in non-small cell lung cancer. Discuss the effection them may cause in development of non-small cell lung cancer.Methods:Carcinoma tissues and corresponding paracancerous tissues (lung tissue less than5cm from the tumor) each80cases were collected from surgical patients who suffered non-small cell lung cancer. These specimens include45cases of lung squamous cell carcinomas,24cases of lung adenocarcinoma and11cases of lung adenosquamous carcinoma;poorly differentiated carcinoma in38cases, moderately differentiated carcinoma in25cases,17cases of well-differentiated carcinoma. Clinical staging:Based on the2009International Union against cancer (UICC) TNM staging criteria,16patients in stage I,1129cases,35cases of stage III. Regional metastatic lymph nodes in34cases and non-metastatic lymph nodes in46cases also were collected. At the same time, normal lung tissue specimens(more than5cm away from the tumor tissue) of52cases were collected as control. FasL, Caspase-8, FADD expression in non small cell lung cancer patients were detected with immunohistochemical S-P method. The results obtained using the13SPSS for corresponding statistical analysis.Results:1. The results of FasL expression in non-small cell lung cancer:There are varying degrees positive expression of FasL in80cases of tumor specimens, FasL is mainly distributed in the cytoplasm, a small part in the cell membrane surface, the positive expression rate is significantly higher than the adjacent tissues positive rate and normal tissues,P<0.05;FasL positive expression rate in lung adenocarcinoma is significantly higher than the positive expression rate in squamous cell carcinoma, and adenosquamous carcinoma,P<0.05; FasL expression positive rate in poorly carcinoma is higher than middle differentiated and well-differentiated carcinoma,there are significant differences(P<0.05).The positive rate of FasL expression in metastasis lymph nodes is higher than in non-metastasis lymph nodes,P<0.05; The difference of FasL expression in different TNM stages in non-small cell lung cancer is not significant (P>0.05),moreover, the relationship between each group of tumor location,ages and sexuality are found no significant difference, no statistical significance (P>0.05).2. The results of Caspase-8expression in non-small cell lung cancer:The distribution of caspase-8is in the cytoplasm,the positive expression rate in cancerous tissue is significantly lower than in tumor-adjacent tissues and normal tissue,P<0.01;and the expression positive rate in lung adenocarcinoma is significantly lower than in lung squamous cell carcinoma and adenosquamous carcinoma (P<0.05); Caspase-8. expression positive rate in well-differentiated carcinoma is higher than middle differentiated and poorly carcinoma,there are significant differences (P<0.05).Caspase-8expression in metastatic lymph group is lower than non-metastatic lymph node group,the difference is statistically significant (P<0.05);Similarly,the differences of caspase-8expression in non-small cell lung cancer in different TNM stage,tumor location, ages and sexuality has non statistical significance (P>0.05).3. The results of FADD expression in non small cell lung cancer:FADD is mainly distributed in the cytoplasm, in cancer tissues positive expression rate is significantly higher than that of the adjacent tissues and normal tissues, P<0.05; FADD positive expression rate in lung adenocarcinoma is significantly higher than that in squamous cell carcinoma and adenosquamous carcinoma, P<0.05; FADD positive rate in poorly carcinoma is higher than middle differentiated and well-differentiated carcinoma, there is significant difference, P<0.05; metastatic lymph node positive rate of FADD is higher than that of non metastatic lymph node groups positive rate, P<0.05; also, in non small cell lung cancer FADD expression in different TNM stage, tumor site, ages and sexuality has non significant difference, non statistical significance (P>0.05).Conclusion:1. There is FasL expression increased significantly in non-small cell lung cancer, and it can make the tumor cells reverse kill immune active cells, so that the tumor form an local immune amnesty parts and is conducive to tumor development and metastasis; also, this may become an important reason for adenocarcinoma to early metastasis more easily.2. Caspase-8expression decreas significantly in non-small cell lung cancer tissue, and the expression in squamous carcinoma is higher than other organization type,which may be the reason that the prognosis in squamous cell carcinoma is better than in adenocarcinoma; and it may become an important indicator to judge the prognosis of non-small cell lung cancer.3. The expression level of FADD increase significantly in non small cell lung cancer tissue, but have nothing with different TNM stage,tumor location, ages and sexuality.
