The Impact of Expression of Dicer and Drosha with Insulin and Hypoxia in Pancreatic Cancer and Its Clinical Significance

ObjectiveTo investigating the regulation of miRNA key enzyme Dicer and Drosha expressionand its expression correlated with clinical in pancreatic cancer; explore the possible reasonsof Dicer and Drosha abnormal expression in pancreatic cancer.MethodsWe select32cases of pancreatic cancer of Institute of Pancreatic Surgery, WuhanUnion hospital from March to November in2012, which have pared adjacentnon-cancerous tissues. Using of fluorescence quantitative PCR detect Dicer and Droshaexpression in pancreatic cancer tissues and adjacent non-cancerous tissues, analyzing Dicerand Drosha expression correlated with pancreatic cancer clinical indicators. Using offluorescence quantitative PCR detect Dicer and Drosha expression in pancreatic cancer celllines with high insulin level or hypoxia enviroment. Using of SPASS19.0statistical andanalysis data.ResultsIn32cases of paired samples，81.3%(26cases) of Dicer mRNA expression wasincreased and78.1%(25cases) of Drosha mRNA expression was increased. The differenceof pancreatic cancer tissue and adjacent non-cancerous tissue expression was statisticallysignificant (P <0.05),75%(24cases) Dicer and Drosha mRNA expression was both raised.Both Dicer and Drosha correlated with pancreatic cancer clinical stage, TNM stage, tumordifferentiation and lymph node metastasis, but has nothing to do with the age and sex of thepatients with pancreatic cancer. Drosha mRNA expression is also associated with tumorsize, Dicer is not relevant. Dicer and Drosha, the higher the level of mRNA expression ofpancreatic cancer correlated with the higher stage of TNM, the lower the degree ofdifferentiation, the more prone to lymph node metastasis; The survival time of patientsbetween Dicer and Drosha mRNA upregulation and downregulation has statisticallysignificance；Insulin can lead to pancreatic cancer cell line Dicer mRNA up-regulation and positively correlated with the concentration and time while Drosha mRNA expression is theopposite; Hypoxic can cause pancreatic cancer cell line Drosha mRNA up-regulation andpositively correlated with the processing time, while the Dicer is the opposite.ConclusionsThe expression of the miRNA and iRNA key enzyme Dicer and Drosha increased inpancreatic cancer. Expression of Dicer and Drosha correlated with pancreatic cancerclinical stage, TNM stage, differentiation and lymph node metastasis, but has nothing todo with the age and sex of the patients with pancreatic cancer. Dicer and Drosha mRNAupregulation is related with the survival time of patients, and may indicate a poorprognosis. Dicer mRNA up-regulation may be correlated with Pancreatic cancer highinsulin environment, and Drosha mRNA up-regulation may be associated with pancreaticcancer hypoxic environment.
