Objective:To investigate the relationship between alumina ceramic particles and asepticinflammation and how lanthanum chloride affect this process through the observationof lanthanum chloride on alumina ceramic particles induced macrophage RAW264.7expressing cytokines(IL-1βand TNF-α). Give a theoretical and experimental basis forthe usage of lanthanum chloride of prevention and treatment of aseptic loosening ofjoint prosthesis.Methods:RAW264.7cells were cultured in vitro and divided randomly into four largegroups(eight small groups)according to different culture solutions: blank controlgroup (group A);alumina ceramic particles group (group B); alumina ceramicparticles+different concentrations of lanthanum chloride(gourp C2.5μmol/L, gourpD10μmol/L, gourp E100μmol/L); different concentrations of lanthanumchloride(gourp F2.5μmol/L, gourp G10μmol/L, gourp H100μmol/L); Detect thecell growth situation by MTT and using ELISA、RT-PCR、western blot to test theexpressions of IL-1β、TNF-αand NF-κb at the level of gene and protein.Results:There is no significantly difference among all groups by MTT(P>0.05).Compared with blank control group (group A),the expressions of mRNA and proteinof IL-1β、TNF-αare significantly higher(P<0.05) in group B and group H,but thereis no significantly difference in group F and group G(P>0.05); When put in thelanthanum chloride, compared with alumina ceramic particles group (group B),theexpressions of mRNA and protein of IL-1β、TNF-αare significantly lower(P<0.05)in group B and group H,but there is no significantly difference in group E (P>0.05);The expressions of mRNA and protein of IL-1β、TNF-αare significantly lower (P<0.05) in Blank control group (group A)、2.5μmol/L lanthanum chloride group (groupF)and10μmol/L lanthanum chloride group (group G) than in100μmol/L lanthanumchloride group (group H); Besides, we tested the NF-κB at the level of gene and protein and the results is similar with above-mentioned.Conclusion:(1)Alumina ceramic particles was little cytotoxic to RAW264.7cells,but it canstimulate the excretion of IL-1βand TNF-αand the expression of NF-κB.(2) Acertain concentration of lanthanum chloride can inhibit the excretion of IL-1βandTNF-α:2.5μmol/L and10μmol/L lanthanum chloride can inhibit the expressions ofinflammatory factors but high concentration of lanthanum chloride (100μmol/L)willeven increase the secretion of the inflammatory factors.(3)The inhibition ofinflammation may be related to the inhibition of NF-κB signal transduction pathwaycaused by lanthanum chloride.
