Tomorrow impact studies on leaf chalcone mouse hepatoma cells Caspase-3 and Bax protein expression

ObjectiveTo investigate the effect of Angelica keiskei Chalcone on Caspase-3 and Bax protein expression and the influence of Angelica keiskei Chalcone on the proliferation of lymphocytes, TNF-a in mice hepatocarcinoma cells.MethodsFifty mice inoculated hepatocarcinoma 22 cells were divided into five groups,10 mice per group.5,20,40mg/kg AC were given daily by mouth to low, medium and high dose group, normal saline by mouth were given to the tumor control group.20mg/kg cytoxan(CTX) by injection every other day were given to the CTX group.10 days later, all mice were sacrificed. The levels of the Caspase-3 and Bax protein expression were measured by immunohistochemistry method, and the proliferation activity of hepatocarcinoma cells and lymphocytes’activity was determined by MTT assay.Use the ELISA method to measure TNF-a.ResultsThe expression level of Caspase-3 and Bax protein in tumor control group were 5.00% and 4.68%, respectively, and those of the high-dose group were 38.52% and 35.76%. The differences between two groups were significant (p<0.05). The cell proliferation activity of tumor control group and high-dose group were 1.135±0.032 and 0.716±0.018.The difference was significant (p<0.05). The proliferation of lymphocytes of tumor control group and high-dose group were 0.278±0.041 and 0.665±0.050,the differences between two groups were significant (p<0.05). The TNF-a of tumor control group and high-dose group were 0.270±0.018 and 0.505±0.077,the difference was significant (p<0.05).ConclusionAngelica keiskei Chalcone can trigger the expression of Caspase-3 and Bax protein, and inhibit the proliferative activity of mice hepatocarcinoma cells.Angelica keiskei Chalcone can increase the immune function of hepatic cancer mice.
