The Study of Lead Exposure on Cyprinus Carpio Ovary Epithelial Cells

Pb contamination was one of the most serious environment pollutions and had a toxic effect on freshwaterfish. Many heavy metals went into the fish body andenriched in the food chain, and then disturbed normal vitalactivity of fish, there by making fish poisonous and dead,thus seriously decrease their output and finally do harmful to peoples’health. However, the toxicity mechanism underlying effects of heavy metal on freshwater species remained largely unclear. But it caused a wide range of biological and biochemical consequences such as enzyme inhibition,fidelity of DNA synthesis, mutation, chromosome aberration, cancer and birth defects.To study the effect on freshwater fish,this study we use cyprinus carpio ovary epithelial cells by lead acetate exposured and we have set three toxic does groups:the high does groups, the medium dose groups,the low does groups and control groups.set two time does groups that are 24h and 48h. we use some technical means including determination antioxidant enzyme, determination lipid peroxid, TUNEL dyeing and Annexin V-FITC PI di- dyeing, Analyze the gray scale of proudct of PCR to study the activition of oxido-reductase enzyme, cells apoptosis, expression of apoptosis genes .First systemic to study and research the effcet of CO afer lead aceate expoursed from cellular level and molecular level. the results showed:1 lead acetate could inhibited the activity of CAT、SOD、GPX、GR enzymes with raised poisonous dose and time dose.And every data are distinguished lower than contorl groups(P<0.05).the levels of MDA was increased. The difference significantly that compared with the control groups(P<0.05).the study showed lead acetate could induced oxidative damage to organism.2 Using the MTT method test showed that lead acetate can suppressed carp oarian epithelial cell proliferation, and with the increase of poisonous dose and time dose, the proliferation inhibition more significantly (P <0.05). Through observated the cell morphology that can see significant apoptosis features, and with the increase of doses and time the more obvious and its characteristics. Adopt TUNEL dyeing and Annexin V - FITC PI double dyeing method can detect the apoptosis phenomenon of carp oarian epithelial cells. and with the increase of poisonous dose and time the number of apoptotic cells also increase. Results show that lead acetate inhibits cell proliferation, and causes cell apoptosis .3 lead acetate exposured, through the method of RT-PCR and analysis the grays of PCR products ,that the expression of inhibiting apoptosis genes Bcl-2 have the reverse effect with poisonous dose and time does.The expression of Bcl-2 was taper with the increase of poisonous doses and time does. Results show that: lead acetate can cause the expression of cell apoptosis gene Bcl-2 decline.
