Study on Preparation of the Injection Clindamycin Phosphate Liposome

Objective:To prepare the clindamycin phosphate liposome with fine appearance, even particle size, high encapsulation efficiency, well stability and lower toxicityMethod:In this paper, the rotation evaporation ultrasound method was used for the loading of clindamycin phosphate into liposome. Using entrapment efficiency as the optimized indexes. The preparation technical parameter was screened by four factors orthogonal analysis method. Its four factor mainly include:the content of cholester, the content of clindamycin phosphate, the concentration of phosphate buffer solution and the water bath temperature. Finally determined the optimum process. A ultra-violet spectrophotometry method was used for the determination of clindamycin phosphate concentration in liposomal clindamycin phosphate and was validated.A method for the determination of entrapment efficiency of liposomal clindamycin phosphate by dialysis sampling-ultra-violet spectrophotometry was established. The physicochemical properties of liposomal clindamycin phosphate were studied. The content of clindamycin phosphate, pH value, particle size and entrapment efficiency have been measured.Results:A method for the determination of entrapment efficiency of liposomal clindamycin phosphate by dialysis sampling-ultra-violet spectrophotometry was established. The results showed that when using rotation evaporation ultrasound method to prepare liposomal clindamycin phosphate, The factor with water bath temperature had the greatest effect to entrapment efficiency,had a range of 39.27%. Next the concentration of phosphate buffer solution, had a range of 15.45%. Once more the content of clindamycin phosphate, had a range of 8.27%. Last the content of cholester, had a range of 4.85%. The prepared liposomal clindamycin phosphate showed little acidity with the diameter of (270.2±41.2) nm. When the weight ratio of cholester to clindamycin phosphate was 5:1, the pH value of phosphate buffer solution was 7 and the water bath temperature was 50℃, the preparation condition is optimal,the prepared liposomal clindamycin phosphate at a pH of about 6.5 and possessed encapsulation efficiency of 61.1%.Conclusion:The optimum conditions of using rotation evaporation ultrasound method to prepare liposomal clindamycin phosphate was obtained through four factors orthogonal analysis. Prepared the clindamycin phosphate liposome with high encapsulation efficiency; small particle size and lower toxicity.
