Adrenomedullin Gene Transfected Bone Marrow Mesenchymal Stem Reduce Apoptosis under Hypoxia-cultured Conditions

[ Objective] adrenomedullin (ADM) gene transfection on bone marrow mesenchymal stem cells (MSCs) proliferation under conditions of hypoxia and serum-free culture , apoptosis , and secretion of vascular endothelial growth factor (VEGF) . [ ] Adherent method vitro isolated, amplified cultured rat bone marrow mesenchymal stem cells . Measured by x-gal staining with adrenomedullin gene recombinant adenovirus (Ad-ADM) , the rate of infection of the bone marrow mesenchymal stem cells . Bone marrow mesenchymal stem cells into untransfected , empty vector Ad -ADM transfected group . In hypoxia, bone marrow mesenchymal stem cells cultured under serum-free conditions in the anoxic 0,3,6,9,12,16,20,24 h CCK - 8 to detect cell proliferation , ELISA detection to detect cell supernatant adrenal adrenomedullin and vascular endothelial growth factor in Tunel assay apoptosis . [ Results ] recombinant adenoviral transfection efficiency of bone marrow mesenchymal stem cells and virus multiplicity of infection (MOI) dose-effect relationship , MOI 150 cell infection rate of 95.4% . Hypoxia , serum-free culture conditions , the three groups of bone marrow mesenchymal stem cells growth inhibition, apoptosis increased , but the adrenomedullin gene transfection group 0,3,6,9,12,16 h apoptosis rate significantly lower than the other two groups ( P lt; 0.05 ) . Hypoxia 9,12 h, bone marrow mesenchymal stem cells secrete adrenomedullin and vascular endothelial growth factor peaked , and Ad-ADM transfection group was significantly higher in the other two groups (P lt ; 0.05) . [Conclusion] in hypoxia under serum-free culture conditions (lt; 20h), the the adrenomedullin gene transfection Enhanced bone marrow mesenchymal stem cell anti-apoptotic ability , which may adrenomedullin , increased expression of vascular endothelial growth factor related.
