Objective: To study autophagy and apoptosis mechanisms Curcumin inhibited glioma cell proliferation . Methods: glioma U87 , U251 cell lines using the MTT assay of curcumin on U87 , U251 cells inhibits the proliferation ; the application flow cytometry curcumin U87 , U251 cells cycle and apoptosis of impact ; application MDC staining , transmission electron microscopy to observe the occurrence of curcumin administration U87 cells autophagy and cell ultrastructure changes ; using western blot study curcumin induced the U87 autophagy in the molecular mechanism . Results: Curcumin showed a dose-dependent inhibition of cell proliferation U87, U251 , U251 cells induced apoptosis , but did not induce the U87 apoptosis rate significantly increased . Curcumin through inhibition of PI3K/AKT signaling pathway raised the level of U87 cell autophagy , to promote autophagic death occurred upregulation of specific proteins LC3 and autophagy protein Beclin1 autophagy . U87 cells inhibit autophagy , p53 , p53 inhibitor Pft-α can up the U87 level of autophagy , and synergistic increase in curcumin - induced autophagy . CONCLUSION: Curcumin inhibition of U251 cell growth by inducing apoptosis through inhibition of PI3K/AKT signal conduction pathway induced U87 autophagic cell death occurred . Pft-α raised U87 level of autophagy , and synergistic increase in curcumin - induced autophagy . The autophagic is one of the ways to inhibit tumor growth .
