Objective:To investigate the protection of the traditional chinese medicine(TCM) named bushenhuoxue(BSHX) fa on purified RGCs in steadily high glucose conditions and glucose fluctuated conditions.and to explor the mechanism. Further more.to provide the experimental evidence of the therapy BSHXfa used in treating diabetic retinopathy (DRP).Methods:l.Two monoclonal antibodies.anti-rat SIRP(OX-41)against rat macrophage and antibody against rat Thy-1.1(OX-7) were used to purify and characterize RGCs from rat neonates by means of two-step filtration.2. Purified RGCs were cultured in steadily high glucose conditions-glucose fluctuated conditions and normal control conditions,BSHX medicine-containing serum was added to the culture medium. The Experiment is assigned into normal control group、TCM interventing group、steadily high glucose group、steadily high glucose group intervened with TCM、glucose fluctuated group、glucose fluctuated group intervened with TCM; 3. Detected the lactate dehydrogenase(LDH) content of extracellular fluid by 490-enzyme-labeled instrument;Deteced the content of the glutamate(Glu) and glycine(Gly) in the extracellular fluid with a full-automatic biochemical analyzer. And then the data obtained was treated with SPSS 13.0.Results:1. The morphology of RGCs had obviously pathological changes in glucose fluctuated conditions.the serum with BSHX decoction can conspicuous reduce pathological damage; 2. Compared with normal control group.the leakage of LDH was obviously increased in glucose fluctuated conditions at 72 hours,the leakage of LDH was obviously decreased by TCM intervention both in steadily high glucose conditions at 72 hours and glucose fluctuated conditions at 48 hours and 72 hours 3. The release of Glu was obviously increased in glucose fluctuated conditions at 24 hours.and the release of Glu was obviously decreased by TCM intervention both in steadily high glucose conditions at each period and glucose fluctuated conditions at 24 hours and 72 hours.4.The release of Gly in glucose fluctuated conditions was obviously decreased, BSHX medicine-containning serum had no significant effect on the release of Gly under different conditions of this experiment.Conclusion:1. The morphology of RGCs had obviously pathological changes in glucose fluctuated conditions, this might be the basis of functional changes in DRP, and BSHXfa could conspicuous reduce the pathological damage.2. BSHX fa could stabilize cells’membrane, decreas its permeability, and decreas its Glu release, accordingly it might be one of the important mechanisms of BSHX fa could prevent and cure DRP.3. BSHX medicine-containning serum had no significant effect on the release of Gly in steadily high glouse conditions、glucose fluctuated conditions and normal control conditions。
