Objective To study the effect of lenti-virus-mediated tandem expression of TNF-βon the growth of human colorectal carcinoma transplanted in nude mice.Methods Human colorectal carcinoma HCT116 cells were anabiosissed, cultivated and passaged enough.And then HCT116 cells were implanted subcutaneously into nude mice. The model of transplanted colorectal HCT116 cell line of colorectal carcinoma in nude mice was set up. Twenty one tumorigenesis nude mice were separated into three groups randomly:The first group is Lenti-virus vector with TNF-βin tandem expression group(Lenti-TNF-βgroup); the second group is Lenti-virus vector group(Lenti-virus group); the third group is normal sodium group(NS group).The given dose corresponding interferent(The Lenti-TNF-βgroup was injected Lenti-virus vector with TNF-βin tandem expression; the Lenti-virus group was injected Lenti-virus; the NS group was injected normal sodium.) was injected into each nude mouse. The transplanted colorectal carcinoma volume and weight were observed. In order to find out the difference, we analyzed the data of the transplanted colorectal carcinoma volume, weight, growth rate and inhibition rate.Results The mean volume of Lenti-TNF-βgroup, Lenti-virus group and NS group was(777.34±63.03)mm3、(1440.81±155.39)mm3 and (1648.62±190.56) mm3; the mean growth rate was (2.38±0.56)、(4.58±0.94) and (5.05±0.48); the mean weight was0.49±0.13) g、(0.78±0.22) g and (1.05±0.32) g, respectively. The Lenti-TNF-βgroup was different from Lenti group and NS group in the mean volume, the mean growth rate and the mean weight(P<0.05). There was no significant difference between Lenti group and NS group in the mean volume, the mean growth rate and the mean weight(P>0.05). The growth of transplanted colorectal carcinoma was significantly inhibited in the Lenti-TNF-βgroup, and the inhibition rate was 44.38%.The inhibition rate of the Lenti-TNF-βgroup was higher than that of the Lenti group and the NS group.Conclusion Lentivirus-mediated tandem expression of TNF-βcan obviously inhibit the growth of human colorectal carcinoma transplanted in nude mice.
