Preliminary Study on the Relationship between Non-small Cell Lung Epithelial Mesenchymal Transition and Metastasis and Its Influencing Factors

Objective The expression of E-cadherin, Vimentin, TGF-β1 and Slug in non-small cell lung cancer (NSCLC) primary tumor tissues and corresponding metastatic carcinoma of lymph nodes were detected to disscuss the relationship between their expressions and clincopathological characteristics. To explore the role of E-cadherin, Vimentin, TGF-β1 and Slug in epithelial mesenchymal transition in NSCLC is in order to provide the basis for the metastasis and prognosis of NSCLC and provide new targets for molecular therapy of NSCLC.Methods①The expressions of E-cadherin, Vimentin, TGF-β1 and Slug were detected by SP immunohistochemical method in 80 NSCLC tumors and 30 corresponding metastatic carcinoma;②Immunohistochemical results were analyzed using statistical software on SPSS 16.0 by means of Mann-Whitney Test for ranked data and Spearman rank correlation test for correlation analysis, P<0.05 for the difference was statistically significant.Results①Compared with corresponding metastatic carcinoma in lymph nodes, E-cadherin,TGF-β1, and Slug expression strength had statistic significance(P<0.05) in the NSCLC tissues. Vimentin expression strength had no statistic significance (P>0.05) between the NSCLC and corresponding metastatic carcinoma in lymph nodes.②The positive expression of E-cadherin,Slug TGF-β1 and Vimentin proteins was closely correlated with TNM stage, differentiation and metastasis of lymph node (p<0.05),but not related to gender, age and histological types(P>0.05).③In NSCLC, The expression of E-cadherin was negatively correlated with that of Slug,TGF-β1 and Vimentin (r=-0.228, r=-0.342, r=-0.226, p<0.05); Slug and Vimentin protein expression was no correlation (r=0.112, P>0.05); the expression of Slug was positively correlated with that of TGF-β1 (r=0.235, P<0.05); Vimentin and TGF-β1 protein expression was positively correlated (r=0.245, P<0.05).Conclusions①It is suggested that EMT phenomenon existed in the growth process of NSCLC. E-cadherin expression was significantly reduced, and Vimentin, TGF-β1 and Slug expression was significantly increased in low differentiation, late clinical stage and lymph node metastasis.②In the NSCLC patients, E-cadherin protein expression reduced or missing and the expression of Vimentin, TGF-β1 and Slug higher indicate patients prone to lymph node metastasis.③Slug and TGF-β1 have certain certain promotive effect in the process of EMT in NSCLC.④It needs to be further studied that if the metastasis showed characteristics of mesenchymal-epithelial transition in corresponding metastatic carcinoma in lymph nodes.
