Effect of rhEPO on the Retinal Neurons in Experimental Diabetic Rats

Objective: To study recombinant human erythropoietin hormone (Recombinat human erythropoietin, rhEPO) provide new strategies for clinical and experimental evidence for the possible protective effect of early diabetic rat retinal neurons . Methods: 45 Wistar rats were randomly divided into three groups , the control group , diabetic model group , and rhEPO intervention group , streptozotocin -induced diabetic rat model ; 2.6 weeks after measuring ERGa wave , b -wave , OPs latency and amplitude to observe the analysis of the retinal light microscopy and electron microscopy sections ; 3 . situ terminal transferase labeling (TUNEL) technique observed retinal neuronal apoptosis . Results : the 1.rhEPO intervention group F-ERGa, b, OPs wavelet Op1 Op2 , Op3 crest latent when compared with the normal group, no significant significance ( P gt ; 0.05) compared with diabetic model group , there were significant ( P lt; 0.05 ) ; the rhEPO intervention group F - ERGa b volatility and OPs total volatility diabetes model group was significant ( P lt; 0.05 ) ; light microscopy , electron microscopy : early diabetic rat retinal nerve ganglion cell layer , inner granular layer structural abnormalities of the retina of diabetic rats of rhEPO intervention group structure near normal ; 3.TUNEL staining : diabetic model group significantly increased apoptosis of rat retinal cells , ganglion cell layer and inner granular layer were seen in a large number of expression of apoptotic cells positive for EPO intervention group retina of diabetic rats apoptotic cells significantly compared to the diabetic group decreased, the difference is statistically significant ( P lt; 0 , 05) . Conclusion : The early diabetic rat retina ERG significant change ; 2 . Retinal neurons undergo apoptosis , diabetic retinopathy retinal ganglion cells and within the external granular layer neuron death one of the important ways ; 3.rhEPO pair apoptosis in early diabetic rat retinal neurons significantly inhibited , and played the role to protect retinal neurons .
