Objective: ① observed Uygur medicine basil alcohol extract in vitro and in vivo cyclooxygenase -1 (COX-1) and cyclooxygenase -2 (COX-2) activity , and to explore the basil with epoxy the dismutase activity relationship to provide a theoretical basis , in order to further explore the mechanisms dimensional antithrombotic drugs basil . ( 2 ) by exploring the dimensional drug adverse reactions of the oral administration of basil , to provide a theoretical basis for the clinical Prescription drug selection . Method : ① vitro : cultured human umbilical vein endothelial cells (ECV304 cell line) and mouse peritoneal macrophages (raw246.7 cell lines ) , determined by MTT cell growth cycle and concentration ; take advantage of the cytotoxicity test basil alcohol extract of dose screening; ECV304 cells were measured by ELISA 6 - keto - prostaglandin F1α ( 6 - keto -PGF1α ) , bacterial lipopolysaccharide (LPS) -induced raw246.7 cells to produce prostaglandin E2 (PGE2) content . ② vivo experiments : with anhydrous ethanol gavage rat gastric ulcer model , observe the rat gastric mucosa , calculate the ulcer index , and the determination of the content of the gastric mucosa 6-keto-PGF1α and PGE2 . Results: ① basil alcohol extracts in vitro inhibition of COX-1 and COX-2 have different degrees of 6 - keto -PGF1α of PGE2 was significantly decreased ( P lt ; 0 , 01) , the same as in the dose , basil alcohol extract on COX-2 inhibition rate than COX-1 (P lt; 0.05). ② basil healthy rat gastric mucosa damage can reduce the ethanol - induced gastric mucosa injury , and make the content of 6-keto-PGF1α increased ( P lt ; 0.05) and no significant inhibition of PGE2 . Conclusion : ① The experiments confirmed basil alcohol extract on COX-1 and COX-2 inhibitory effect , Mechanism of antithrombotic part with likely due to inhibition of cyclooxygenase -mediated 's . ② basil of normal gastric mucosa damage can reduce the area of ethanol - induced gastric ulcer , with better protective effect on the gastric mucosa , can be widely used set of square .
