Salicornia herbacea L. is a kind of halophytes introduced from Europe and America, which contains many nutrients and active ingredients with anti-oxidative, anti-microbial, anti-proliferative, and anti-inflammatory activities. This plant has been reported to be effective on cancer, diabetes and hepatitis. Unfortunately there is very little information available concerning the active ingredients and physical activity of Salicornia herbacea in our country, and most studies focused on its introduction, as well as cultivation and salt-tolerance mechanism.This paper presents investigations on the extraction, separation and purification of fat-soluble constituents from Salicornia herbacea powder, and the biological activity of the fat-soluble constituents was also investigated. The aim of this study was to provide a biologically active substances of natural origin and theoretical basis for further development for the utilize of Salicornia herbacea L. The results were as follows:1. The solvent leaching method was selected to extract fat-soluble active constituents from Salicornia herbacea in this paper. In order to optimize extraction condition, residue after water extraction as raw materials and ethanol as solvent extraction raw materials and extraction solvent were screened by a guided 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging assay. Through single factor experiments and orthogonal test, the optimal extraction conditions was 3h extraction time,75℃extraction temperature,1:20 solid-liquid ratio,3 times extraction times. The extraction rate was 5.14%.2. To study further the properties of the fat-soluble active ingredients of Salicornia herbacea, separation and purification of the natural antioxidant ingredients was investigated. Two active ingredients can be obtained by ethyl acetate extraction, silica gel column chromatography, Sephadex LH-20 gel filtration chromatography and and high performance liquid chromatography. The molecular weight of this active ingredient was 802 by LC-MC. The silica gel column was gradient eluted with chloroform-methanol=10:0、8:2、6:4、4:6、2:8、0:10 (V/V), constant gradient eluted with 100% methanol in the process of SephadexLH-20. All fractions were detected at UV 225nm using a UV spectrophotometric.3. In vitro, antioxidant activity of Salicornia herbacea ethyl acetate fractions (SEF) were studied. There are scavenging ability of SFE on the hydroxyl radical, DPPH and reducing power, but less than the positive control Vc. SFE had a certain inhibition of lipid peroxidation, in the concentration of less than 1mg/mL, the inhibition was greater than Vc, the concentration of more than lmg/mL, the inhibition was less than Vc.
