The Research of Kaempferol to Interfere with Hepatic Fibrosis Process Which Results from Schistosomal Granulomas

Objecitve: To observe effectiveness of hepatic fibrosis mice with experimental schistosomiasis treated by kaempferol and it’s possible correlate antifibrotic mechanism of action through the experiment.Methods: Through the method of infecting the skin of mouse abdomen by Japanese schistosome cercarias , to make mouse model of schistosomiasis hepatic fibrosis. In the experiment one , infected Kunming mice were randomly divided into 4 groups: mice in Chinese medicine treatment group were lavaged by kaempferol; mice in praziquantel treatment group were lavaged by praziquantel; mice in model control group were lavaged by Sodium Chloride; mice in normal control group were not infected by schistosomiasis. Mice in every group were lavaged from the sixth week. Observed the hepatic fibrosis of every group through HE dyeing and Masson dyeing. In the experiment two, uesd the praziquantel to kill the Balb/c mice’s insect after six weeks and then mice were randonmly divided into kaempferol treatment group, model group and normal group according to the condition of use different medicine respectively. The content of hydroxyproline in liver homogenate, the levels of IL-4, IL-13 in liver homogenate, the content of Arg-Ⅰin liver homogenate were assayed by alkaline hydrolysis, ELISA and Real-time quantitative PCR method, respectively. We could identify different expression condition of various kinds of materia between 3 groups through above methods.Result: Compared with praziquantel treatment group, we found that the Kunming mice infected Japanese schistosome were treated by Chinese medicine kaempferol, whose liver of schistosoma ovum nodules didn’t decrease through the screening in the experiment one , but the degree of hepatic fibrosis significantly reduced(P<0.05); compared with model group ,the mice’s degree of hepatic fibrosis obviously reduced in kaempferol treatment group (P<0.05). Compared with model group ,the contents of hydroxyproline, IL-4, IL-13 and Arg-Ⅰin liver homogenate all significantly reduced in kaempferol treatment group(P<0.05).Discuss: 1.Kaempferol can significantly inhibit schistosomiasis ovum granuloma and hepatic fibrosis process. It has intervene efficacy in Japanese schistosomiasis hepatic fibrosis. 2. Kaempferol may down-regulate the expression of IL - 4, IL - 13 and Arg–Ⅰ, inhibit the degradation of liver hyp so as to prevent schistosomiasis hepatic fibrosis .
