The Protective Effect of Rat Myocardial Ischemia/reperfusion Injury Following Bone Marrow Mesenchymal Stem Cell Pretransplantation for 1 Week

BACKGROUND: Acute myocardial infarction with thrombolysis or open blocked blood vessels involved in means,OBJECTIVE: To study the method of isolated culture of bone marrow mesenchymal stem cells(BMSCs), and to observe the effect of BMSCs to the Myocardial ischemia-repefusion injury.METHODS: BMSCs were cultured and expanded from healthy 3-week-old SPF mice in vitro. 48 recipient SD rats were randomly divided into four groups(n=12):The control, sham, ischemia-reperfusion group and BMSCs injection group. BMSCs were transplanted into BMSCs group mice and DMEM was injected into IR group and sham group mice. The Myocardial ischemia-reperfusion injury(IR) model was established seven days later.serum lactate dehydrogenase(LDH) activity,total superoxide dismutase (TSOD) vitality and malondialdehyde (MDA) of myocardial tissue were measured. We examined the changes in the myocardium ultrastructure resulting from coronary ischemia-reperfusion . Cell apoptotic index (AI) was examined by means of terminal deoxynucleotidyl transferase-mediated d-UTP nick end labeling (TUNEL). The expressions of Bcl-2 and Bax proteins were measured by immunohistochemical technique.RESULTS AND CONCLUSION: BMSCs not only had a high purification, homogeneity and stability, but also had unlimited proliferation. BMSCs group compared with IR group, LDH activity, TSOD activity and MDA levels were statistically significant differences (P<0.01). myocardial ultrastructure injury was decreased.The apoptosis index was 21.4% in BMSCs group, which was significantly decreased compared with that in IR group 39.2%(P <0.01).The expressions of Bcl-2 in Group BMSCs were more than those in Group IR(P <0.05), and the Bax/Bcl-2 ratio in Group BMSCs were fewer than those in Group IR(P<0.05).BMSCs transplantation can promote cells proliferation of the Myocardial ischemia-repefusion injury.
