The Changes of Intimal Macrophages, Smooth Muscle Cells, Collagen, and Cpt-Ⅰ, GLUT-4 of Vascular Media in Human Lower Limb Artery with Atherosclerotic Stenosis

Objective To investigate variations of intimal thickness, macrophages, collagen, and CPT-I, GLUT-4 of vascular media in human lower limb with atherosclerotic stenosis.Methods To collect the proximal and distal artery of patients with lower limb atherosclerotic stenosis and amputation. Then took artery and fixed for paraffin sections. Paraffin sections were stained by hematoxylin-eosin and picro-siriusred and immunohistochemistry. Intimal thickness, macrophages, smooth muscle cells, collagen, and CPT-I, GLUT-4 of media were studied separately.Results 1. In the proximate of atherosclerotic stenosis, the intimal thickness of was thicker than that of the distance. [(450.82±193.90 vs 372.81±172.67)μm, P<0.05].2. In the proximate of atherosclerotic stenosis, macrophages and collagen were significantly more than that of the distance. [(134.89±21.65 vs 112.66±14.67)IOD, (152.25±42.55 vs 139.81±27.61) IOD, P<0.05]3. In the proximate of atherosclerotic stenosis,the expression ofα- actin and GLUT-4 of media was significantly higher than that of the distance.[ (162.94±42.67 vs 143.38±36.90)IOD,(141.13±28.01 vs 130.47±32.19)IOD, P<0.05].4. Comparing with the distance of atherosclerotic stenosis, the expression of CPT-Ⅰof media reduced in the proximate. [(139.09±25.94 vs 148.72±26.66)IOD, P<0.05].Conclusion In human lower limb with atherosclerotic stenosis, the intima of proximate was thicker compared with the distance; Intimal macrophages, smooth muscle cells and collagen of the proximate increased compared with the distance; In the proximate of atherosclerotic stenosis, GLUT-4 of media increased compared with the distance, but CPT-Ⅰdecreased.
