Therapeutical Effects of Diammonium Glycyrrhizinate Lipid Ligand on Nonalcoholic Steatohepatitis and Its Partly Mechanisms of Action

Nonalcoholic fatty liver disease (NAFLD) is an emerging worldwide common problem,with all unclear nature history that is frequently associated with obesity, hyperlipidemia and type 2 diabetes mellitus.This disease,can be severe and is characterized by a wide pathological lesions.Nonalcoholic steatohepatitis (NASH) showes hepatic steatosis, ballooning degeneration, diffuse hepatic lobule inflammation and (or) liver next to a central vein, liver sinus around features of collagen deposition of chronic liver disease. NASH often progresses to more severe stages of liver injure,such as fibrosis,cirrhosis and even hepatocellular carcinoma.Though NASH may proceed into end-stage liver disease, the optimal treatment has not been established,which has used adjustable lipid medicine combined liver cells protectants or NASH antioxidants, but the safety and efficacy was further evaluation.Diammonium glycyrrhizinate lipid ligand (DGLL) (Tianqing GanPing) is the active ingredients of liquorice extract 3rd generation, have stronger anti-inflammatory, protective liver cell membranes and improve liver function. The clinical drug used to treat various fatty liver, DGLL is 18-alpha glycyrrhizic acid and ammonium with soy lecithin formation of lipid ligand, its resistance to liver damage effect obviously superior DGLL glycyrrhizic acid.PURPOSE The DGLL treating nonalcoholic function and mechanism are rare reports at present. The aim of the present study is to evaluate the therapeutical effects of DGLL on NASH and to analyze possible mechanisms of DGLL effects in rats fed with high fet diet.METHOD Establishing NASH rats model, analyzing the pathological and histological impact of NASH rat liver infected by DGLL by light and electron microscope; observing the changes of TNF-α, NF-κB p65 protein, I-κBαproteins and CYP2E1 expression level of NASH rat liver tissue by Western Blot and immunohistochemical method to investigate the influence to rat inflammation by DGLL; Examining the changes of SOD activity, GSH, MDA content in liver tissue and ALT、AST、TC、TG、HDL、LDL of rat serum to observe the influence of oxidative metabolism in NASH rat by DGLL.RESULTS By hepatic histopathological observing, DGLL can improve hepatic steatosis, inflammation and necrosis degree; DGLL can reduce TNF-αexpression in liver tissue and serum of NASH rat, and decrease the expression of NF-κB p65 protein expression in rat liver tissue and suppressing degradation of I-κBαin liver tissue; DGLL obviously reduce TG, TC, FFA, LDL - C level in rat serum and TG, TC, FFA level in liver tissue, and increase HDL-C level in rat serum as well.CONCLUSION DGLL has obvious function in the treatment to NASH rats, and its function mechanism has relationship with lipid metabolism, antioxidation function and inflammation reduction.
