The Expression of MMP-2、 VEGF and Ki-67 and Human Glioma Grade

Objective: Human glioma is the most common brain tumour in the intracalvarium, Because of its biological characteristic of diffuse infiltration into neighbouring brain tissue, it often trended to recur. It is difficult to treat and achieve the goal of curing. To explore the expression of MMP-2, VEGF and Ki-67 in human glioma and its correlation with glioma grade . Method: 50.cases of human glioma and 6 cases of normal human tissue were collected from the First and Third Affiliated Hospital Shihezi Medical University(February 2002 to December 2009), study section preparation again. The samples from 50.cases of glioma were divided into four groups according to the WHO classification: gradeⅠ-Ⅱ(n=17), gradeⅢ(n=21), gradeⅣ(n=12), 6 normal brain samples were served as the control. Immunohistochemical method(EnVision)was used to examine the level of MMP-2, VEGF and Ki-67 in surgical samples from 50.patients suffered from glioma and 6 cases of normal brain tissue, and the correlation among the expression of MMP-2, VEGF and Ki-67 and glioma grade was analyzed . Result: The MMP-2, VEGF and Ki-67 were not expressed in all the normal brain tissues, but the expression of MMP-2, VEGF and Ki-67 could be detected in gradeⅠ-Ⅱ,Ⅲand,Ⅳof glioma, there was significant difference in the expression of MMP-2, VEGF and Ki-67 among the normal brain tissue group, the low grade glioma group and the high grade glioma group(Р<0.01),With the increase of the pathological degrees the MMP-2 and VEGF expression increased too, there was a significant Positive correlation(χ2=16. 82, P<0.001)among the expression of MMP-2, VEGF and Ki-67 in all the pathological grades of gliomas . Conclusion: (1) The expression of MMP-2、VEGF and Ki-67 has positive correlation to the glioma grading.It implied that the high expression of MMP-2、VEGF and Ki-67 protein might make great contribution to cellular differenation and malignant progress.(2) There was a positive correlation among the expression of MMP-2、VEGF and Ki-67 protein.It implied that the expression of MMP-2、VEGF and Ki-67 protein had the consisteney. MMP-2、VEGF and Ki-67 play cooperative roles in the occurrence and malignant development of the glioma.(3) The united detection of MMP-2、VEGF and Ki-67 may increase the diagnostic accuracy of the glioma and guide the clinical treatment. The expression of MMP-2, VEGF and Ki-67 was three objective biological markers for estimating the glioma grade .
