Expression of MMP-9 and p63 in Oral Squamous Cell Carcinoma and Clinical Significance

Background:Oral squamous cell carcinoma is one of the most fatal cancers in our country nowadays ,Its incidence has a rising trend in the world .According to the interrelated data in our country , oral cancer accounts for 1.9% - 3.5% in systemic malignant tumor ,14.7% - 20.3% in head and neck malignancy.Oral and maxillofacial has rich in blood vessels and lymph nodes , oral cancer can early be transported by blood supply or lymph node ;sometimes the primary lesion is little or no obvious symptoms , but metastasis can be foundIn the neck. Therefore the mortality rate is high, oral cancer prognosis also is not ideal.With the development of biochemistry, immunology and molecular biology, cell engineering and genetic engineering corresponding detection technology, according to tumor markers existing in patient’s blood, urine or other body fluids , we can not only assist in the diagnosis of tumour , but also in the effective prevention , prognosis and adjuvant therapy.Objective:This study was designed to investigate the protein expression of MMP-9 and p63 in the progression of oral squamous cell carcinoma from protein molecular level. and we also analyse the relationship of MMP-9 and p63 with patients’the clinical and pathological features ,such as age, sex, tumor size, lymph node metastasis and tumor cell differentiation degree, in order to direct the clinical work effectively.Methods:In this study, 68 cases of oral squamous cell carcinoma patients were selected as the the case group, 68 cases of tumor-adjacent normal tissues for the control group. Immunohistochemical methods were used to investigate the expression of MMP-9 and p63 in 68 cases with oral squamous cell carcinoma and 68 cases with normal tissues in an effort to obtaining data. All samples must follow the standards: the first cases, definite diagnosis, no other head and neck cancer history, no radiotherapy and chemotherapy history.Results:MMP-9 and p63 positive expression were detected to a greater or lesser degree in all the cases. Positive expression of MMP-9 were found 76.5%(52/68) in oral squamous cell carcinoma,and positive expression of p63 were found 79.4%(54/68) in oral squamous cell carcinoma ,significantly higher than normal tissues, evidently . The positive expression rate of MMP-9 and p63 in oral squamous cell carcinoma was no relationship with patient’s age, gender and the tumor size. but the positive expression rate of MMP-9 in OSCC increased significantly and correlated closely with the progression of tumor pathological stage.The positive expression rate of p63 in OSCC correlated closely with the progression of tumor differentiation grade.Conclusions:The expression of MMP-9 and p63 in OSCC correlates closely with the invasion and metastasis of OSCC,which will be a new biological markers to predict the prognosis of OSCC.
