Objective: decreased and the weakening of the structure of the extracellular matrix (extracellular matrix, ECM) is a key link to premature rupture of membranes, some studies suggest that matrix metalloproteinases (martix metalloproteinases, MMPs) and their inhibitors TIMP (tissue inhibitor of metalloproteinase) transforming growth factor (transforming growth factor, TGF-β1) plays an important role in the degradation of ECM process of MMPs TIMP and TGF-beta1 in premature rupture of membranes in the mechanism of action is not clear, the purpose of this paper is to explore patients with premature rupture of membranes placenta, fetal membranes in MMP-9, TIMP-1 and TGF-beta1 expression changes and significance. Methods: SP immunohistochemical and Western blot blot hybridization was detected 28 cases of term not in labor elective cesarean section in 28 cases, 28 cases of spontaneous onset of labor at term vaginal delivery and premature rupture of the fetal placenta in pregnant women after childbirth The membrane tissue MMP-9, TIMP-1 and TGF-beta1 expression changes. Results: Immunohistochemical results shown 1 not in labor group fetal membranes in the lowest intensity of MMP-9 expression (P lt; 0.05), followed by labor group MMP-9 expression strength (P lt; 0.05), premature rupture of membranes group MMP-9 expression highest intensity (P lt; 0.05) MMP-9 expression in the placenta trend likewise. ② expression of TIMP-1 and TGF-beta1 expression trends and MMP-9 In contrast, in the fetal membranes, not in labor TIMP-1 and TGF-beta1 expression (P lt; 0.05), the labor group TIMP-1 and TGF-beta1 expression strength followed by (P lt; 0.05), the PROM group TIMP-1 and TGF-beta1 expression lowest (P lt; 0.05), placenta TIMP-1 and TGF-beta1 versa. Western blot blot method results shown (1) not in labor group fetal membranes of MMP-9 was the lowest (P lt; 0.05), followed by labor group MMP-9 expression (P lt; 0.05), premature rupture of membranes group MMP-9 expression (P lt; 0.05), MMP-9 expression trend in the placenta same. (2) is not labor group in the fetal membranes TIMP-1 and TGF-beta1 expression highest (P lt; 0.05) the labor group TIMP-1 and TGF-beta1 expression, followed by (P lt; 0.05), premature rupture of membranes group TIMP-1 and TGF-beta1 was the lowest (P lt; 0.05), placenta TIMP-1 and TGF-beta1 versa. (3) the level of TGF-beta1 expression in placenta and fetal membranes and TIMP-1 expression was positively correlated with MMP-9 expression was negatively correlated. Conclusion: TGF-beta1 expression in placental membranes reduced, may regulate the expression of MMP-9 and TIMP-1, causing MMP-9/TIMP-1 ratio between the imbalance between collagen degradation accelerated fetal membrane strength, toughness decreases , resulting in the occurrence of premature rupture of membranes.
