Study on the Expression and Correlation of Metallothionein and Matrix Metalloproteinase-2 in Non-small Cell Lung Cancer

Objective To investigate the expression correlation and clinical significance of metallothionein(MT) and matrix metalloproteinase -2(MMP-2) in the non-small cell lung cancer(NSCLC). Methods SP immunohistochemistry staining was used to examine the expression of MT and MMP-2 proteins in 71 cases non-small cell lung cancer (NSCLC) tissue,15 cases peritumoral tissue(PT) of non-small cell lung cancer (NSCLC),4 cases pulmonary benign tumor (BT) tissue and 5 cases normal lung tissues (NLT). Leica DM4000B microscope camera was used to capture images of immunohistochemistry sections and Image-Pro Plus 6.0 image analysis software was used to analyze these images, in each case selected a representative tissue section to analyze,randomly selected high power (SP×400) field of vision to observe and capture 20 images in each section. The integral optical density (IOD) value of each image was calculated by Image-Pro Plus 6.0 image analysis software, the mean integral optical density (IOD) value of 20 images was treated as the positive protein expression quantity of MT or MMP-2 proteins of each case. Results (1). Metallothionein (MT) and matrix metalloproteinase -2 (MMP-2) proteins are expressed in normal lung tissue(NLT),peritumoral tissue(PT), benign tumor tissue (BTT) and lung cancer tissue(LCT). (2). The IOD values of MT and MMP-2 expression in CT group are higher than that of NLT group,PT group and BT group, there are statistical significance(p<0.05), in the PT group is higher than NLT group and BT group also, there are statistical significance(p<0.05), but the expression of MT and MMP-2 in NLT group and BT group are no statistical difference (p>0.05). (3). The IOD values of MT and MMP-2 expression in the less than 50 years old group are higher than that of the greater than or equal to 50 yeas old group, there are statistical significance(p<0.05); MT and MMP-2 expression is more correlated in the less than 50 years old group compared to the greater than or equal to 50 yeas old group (r=0.706,p<0.01). (4). The IOD values of MT and MMP-2 expression in male group are higher than that of female group, there are statistical significance(P<0.05); MT and MMP-2 expression is more correlated in male group compared to female group (r= 0.702, p<0.01).(5). The expression of MT and MMP-2 proteins in lung cancer tissue is correlated with histological type, histological grade, tumor TNM stage and lymph node metastasis, but they are not correlated with tumor size(p>0.05). The IOD values of MT and MMP-2 expression in adenocarcinoma group are higher than that of squamous cell carcinoma group, there are statistical significance (p<0.05); MT and MMP-2 expression is more correlated in adenocarcinoma group than that of squamous cell carcinoma group(r= 0.636.p<0.01). The IOD values of MT and MMP-2 expression in poorly differentiated carcinoma group are higher that of compared to moderately differentiated and highly differentiated group, there are statistical significance (p<0.01), in the moderately differentiated carcinoma group are higher than highly differentiated group also, there are statistical significance(p<0.05); MT and MMP-2 expression is more correlated in poorly differentiated group than that of moderately differentiated group and highly differentiated group (r=0.752. p<0.01). The IOD values of MT and MMP-2 expression in TNMⅢstage group are higher than that ofⅠstage andⅡstage group, there are statistical significance(p<0.01), inⅡstage group are higher thanⅠstage group also, there are statistical significance(p<0.05); MT and MMP-2 expression are more correlated in TNMⅢstage group than that ofⅠstage andⅡstage group (r=0.673, p<0.01). The IOD values of MT and MMP-2 expression In lymph node metastasis group are higher that of without lymph node metastasis group, there is statistical significance(p<0.01); MT and MMP-2 expression are more correlated in lymph node metastasis group than that of without lymph node metastasis group (r=0.702. p<0.01). (6). Between expression of MT and MMP-2 proteins is positive correlated in the lung tissue(r=0.481,P<0.01). Conclusion There are correlation between the expression of metallothionein (MT) and matrix metalloproteinase-2 (MMP-2) proteins in non-small cell lung cancer (NSCLC). MT and MMP-2 maybe affect the process of occurrence, growth, invasion and metastasis of NSCLC by their interaction, detected MT and MMP-2 high expression in NSCLC tissues can be considered to determine reliable indicators of NSCLC patients prognosis.
