The dried radix of Juglans cathayensis Dode has been used as a folk medicine for the treatment of breast cancer, stomach pain and esophageal cancer in China. Screening of antitumor activity of the radix extraction in vitro suggested that the ethyl acetate and n-butanol fractions were the active parts.On this basis, the ethyl acetate fraction was separated systematically and 29 compounds were isolated. 27 compounds were identified by their physical and chemical constants, spectra data and comparing with the laboratory samples isolated before. They are juglone (YHT-1), 2-methoxy juglone (YHT-2), 3-methoxy juglone (YHT-3), 2-ethoxy juglone (YHT-4), 5,8-dihydroxy-1,4-naphthoquinone (YHT-5), regiolone (YHT-6), Butyric acid, 4-[(1,4-dihydro-5-hydroxy- 1,4-dioxo-2-naphthalenyl)amino]- (YHT-7), [1,2’-Binaphthalene]-1’,4’,4-trione, 1,2,3-pentahydro-,-5,8’,8-trihydroxy-,-3’-amoni- (YHT-8), engelharquinone (YHT-9), 4,8-dihydroxy alpha-naphthol-1-O-β-D- [6′-O-(3″, "5" - dimethoxy- 4– hydroxyl-benzoyl)] glucopyranoside (YHT-10), galleon (YHT-11), 3′4″-epoxy-l-(4′-hydroxyphenyl)-7-(3″-methoxylphenyl)-heptane-2-hydroxy-3-one (YHT-12), 1, 2, 3, 6-tetra-O-galloyl-β-D-glucose (YHT-13), 1, 2, 4, 6-tetra-O-galloyl-β-D-glucose (YHT-14), 1, 2, 6-tri-O-galloyl-β-D-glucose (YHT-15), quercetin (YHT-16), kaempferol (YHT-17), dihydrokaempferol (YHT-18), vanillic acid (YHT-19), gallic acid (YHT-20), caffeic acid ester (YHT-21),β-amyrin (YHT-22), oleanolic acid (YHT-23),β-sitosterol (YHT-24), daucosterol (YHT-25), methyl-O-β-D-glucopyranosid (YHT-26) and n-docosanoic acid (YHT-27) respectively. YHT-7 and YHT-8 are two novel compounds. YHT-9 was isolated from Juglans for the first time. YHT-1,-2,-3,-5,-10,-11,-12,-13,-18,-20,-21,-22,-23,-26,-27 were isolated from this plant for the first time.Through a series of pharmacological screening experiments, we found that there were two kinds of bioactive constituents in Juglans cathayensis Dode. One kind is naphthoquinone compounds which have the antitumor activity; the other kind is gallotannin compounds which have the promoting cell proliferation activity.
