Effect of Hyperlipemia on GFAP and bFGF Expression in the Hippocampus after Cerebral Ischemic-reperfusion Injury

Objective: To observe high cholesterol on cerebral ischemia-reperfusion rat hippocampal GFAP and bFGF expression, to explore the cerebral ischemia-reperfusion injury mechanisms to provide evidence. Methods: normal healthy adult Sprague-Dawley rats 166, weight 160-180g, by the Experimental Animal Center of Shanxi Medical University. Randomized cerebral ischemia-reperfusion group (52), high cholesterol with cerebral ischemia-reperfusion group (52) and sham operation group (62) the three groups. Two sets of ischemia-reperfusion group were further divided into ischemia-reperfusion 2h 1d (I2h/R1d), 3d (I2h/R3d), 7d (I2h/R7d), 14d (I2h/R14) 4 groups of 13 , respectively, for immunohistochemistry, HE staining and Western blot 6 6, each one for TTC staining; sham operation group for immunohistochemistry and HE staining of 48 (high fat sham 24 only the normal sham group 24) for Western blotting 12, 2 for TTC staining. High-fat diet group model uses high fat diet ad libitum production. After six weeks, automatic chemical analyzer with a total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL-L), low density lipoprotein (LDL-L), the high-fat diet former was statistically significant, that model was successfully. On this basis, for cerebral ischemia and reperfusion model. Ischemia-reperfusion group with middle cerebral artery occlusion model making. With neurological and TTC staining, inspection ischemia-reperfusion group model is successful. Results: 1.GFAP and bFGF expression: GFAP and bFGF immunoreactive cells in the hippocampus CA4 region common. Immunoreactive cells in the sham group is weak; cerebral ischemia-reperfusion group I2h/1d increased, I2h/3d peak, I2h/7d down, I2h/14d expression was significantly decreased; GFAP hyperlipemia with cerebral ischemia-reperfusion group peak expression Point postponed, I2h/7d reached the highest, bFGF hyperlipemia with cerebral ischemia-reperfusion group with cerebral ischemia-reperfusion group was the same. Westernblot show the same results. (2) at the same time points, GFAP and bFGF hyperlipemia with cerebral ischemia-reperfusion group and cerebral ischemia-reperfusion group compared to the positive expression was significantly reduced. 3.GFAP and bFGF expression similar trend, correlation analysis showed a positive correlation coefficient of 0.5. Conclusions: 1. Cerebral ischemia and reperfusion, GFAP and bFGF immunoreactive cells in the hippocampal astrocytes CA4 region common. Description GFAP, bFGF may be involved in hippocampal cerebral ischemia-reperfusion injury repair. 2.GFAP and bFGF in two cerebral ischemia-reperfusion group expression was dynamic, and the expression of GFAP and bFGF in time with consistency, correlation analysis showed a positive correlation, suggesting: bFGF on expression of GFAP upregulation. 3 At the same time,. GFAP and bFGF hyperlipemia with cerebral ischemia-reperfusion group and cerebral ischemia-reperfusion group was significantly decreased compared to expression; hyperlipemia with cerebral ischemia-reperfusion group scored significantly higher neurological in cerebral ischemia-reperfusion group, were prompted high cholesterol increased brain ischemia-reperfusion injury.
