Effect and Mechanism of Asiatic Acid on Angiogenesis

Objective: To study the effect and the molecular mechanism of asiatic acid on angiogenesis in Eahy926.Methods: To study the effect of asiatic acid on angiogenesis,we used Annexin V and PI double staining,Hoechst33258 staining and two endothelial cell basal model:(1)migration,(2)tube formation using Eahy926, an endothelial cell line. To study the molecular mechanism of asiatic acid on angiogenesis, we used western blot to detect the level of ERK1/2, p-ERK1/2 , Akt, p-Akt.Results: Results demonstrate that certain concentrations of asiatic acid (25μmol/L、30μmol/L ) induces Eahy926 apoptosis, inhibits Eahy926 migration, inhibits tube fomation, We also found the level of ERK1/2, p-ERK1/2 is decreased by various concentrations (20μmol/L、25μmol/L、30μmol/L ) of asiatic acid in Eahy926.Conclusion:Asiatic acid inhibits Eahy926 migration and tube fomation. Asiatic acid suppresses tube formation in Eahy926 endothelial cell may be associated with Eahy926 apoptosis through inactivation of survival signal ERK1/2.
