The Regulation and Research of Mechanism of "Plus Sijunzi Decoction" in Immune System of the Aging Rat

Objective: Observe the regulation function of traditional Chinese medicine "Plus SiJunZi Decoction" in the immune system of aging rats, study of its anti inflammatory effects of aging, and discusse the preliminary mechanism of action.Methods: (1) Select 50 Wistar (male 25 and female 25) rats of 12 months old, feeding with normal diet adaptive for one week. Randomly divided into 5 groups, "Plus SiJunZi Decoction" (hereinafter referred to as drugs) low dose group, middle dose group and high dose group, positive control group, the elderly control group,10 each group; selected another 3 months of 10 healthy Wistar rats were used as young control group. (2) were given "Plus SiJunZi Decoction" extract 1.6g/kg/d, 3.2g/kg/d, 6.4g/kg/d, "Liu Wei Di Huang Wan" solution 1.5g/kg/d, 2ml of distilled pour stomach, continuous treatment for 90 days. (3) weighing the weight of rats were sacrificed to observe the rats the following indicators: body weight of rats in each group (BW); serum immunoglobulin (IgG) level; calculate the thymus index and spleen index; the thymus and spleen by HE staining, observed morphological changes under light microscope; positioning using immunohistochemistry and quantitative analysis of thymic interleukin-2 (IL-2) expression and spleen of tumor necrosis factorα(TNF-α) expression; use of real-time fluorescence Quantitative PCR method for detection of nuclear factor-κB (NF-κBp56) mRNA expression levels.Results: (1) The changes in body weight of rats in each group: before and after treatment, the rats increased in weight; the drug group weight of rats in each group than the positive control group, the elderly control group no significant difference. (2) The comparison of serum IgG level: compared with the young control group, serum levels of IgG old were significantly lower (P<0.01); compared with the aged control group, IgG levels increased drug group (P<0.01), positive control IgG levels increased (P<0.01); compared with the positive control group, drug group were significantly lower in IgG level of (P<0.05). (3) rats thymus index and spleen index in comparison: compared with the young control group, the older the group, thymus and spleen index were significantly lower (P<0.01); compared with the aged control group, drug group thymus index was significantly higher (P<0.01), spleen index was no significant difference (P>0.05), positive control group thymus index were significantly higher (P<0.01), spleen index was no significant difference (P>0.05); compared with drug group and the positive control group, the thymus index and spleen index were not significant different (P>0.05). (4) Rats thymus, spleen Morphological comparison: Drug group thymus had varying degrees of atrophy, reduce thymus pathological changes is the degree of fat substitution, more frequent distribution of thymus cells, coloring is good. Compared with the aged control group, treatment group spleen white pulp and red pulp of the boundaries less clear, a slight increase in white pulp, increase in the number of neutrophils, lymphocytes arranged loosely. (5) the expression of thymus IL-2 levels: compared with the young control group, the rats aged thymus expression of IL-2 were significantly lower (P<0.01); compared with the aged control group, drug group thymus IL -2 expression was increased (P<0.01), positive control thymic expression of IL-2 levels (P<0.01); compared with drug group and the positive control group, IL-2 expression in the thymus was no significant difference (P>0.05). (6) the expression of splenic TNF-αlevels: compared with the young control group, the rats aged spleen expression of TNF-αwere significantly higher (P<0.01); compared with the aged control group, treatment group spleen TNF-αexpression levels decreased (P<0.01), positive control spleen TNF-αexpression levels decreased (P<0.01); compared with drug group and the positive control group, the spleen TNF-αexpression level of the difference was not significant (P>0.05). (7) the rats the expression of spleen NF-κBP65mRNA: compared with the aged control group, NF-κBp65mRNA young rats was significantly higher (P<0.01); compared with the positive control group, NF-κBp65mRNA was significantly higher (P<0.05), NF-κBp65mRNA drug group was significantly higher, the difference was statistically significant (P<0.05, P<0.01); and the positive control group, low dose of drug increased expression of NF-κBp65mRNA (P>0.05); high dose group was significantly higher, the difference was statistically significant (P<0.01). Conclusion: "Plus SiJunZi Decoction" in the rat during aging applications that can increase the serum IgG levels in aged rats, increased the thymus index, spleen index were not affected, increased thymic expression of IL-2, reduced expression of TNF-αin the spleen, reduced expression of NF-κBP65mRNA spleen tissue, suggesting that "Plus SiJunZi Decoction" possibly through regulation of NF-κB signaling pathway expression, leading to IL-2 expression increased, TNF-αexpression level decreased, increased humoral immunization, control of the high inflammatory status of the body, thereby delaying the inflammatory immune senescence in aging rats.
