Relationship between Endothelial Cell Derived Microparticles and aGVHD in Allogeneic Hematopoietic Stem Cell Transplantation

Purpose by monitoring allogeneic hematopoietic stem cell transplant patients, and patients with peripheral blood endothelial microparticles (EMPS) and endothelial injury Biology media Ang - 1 , Ang -2 , VEGF , TNF -α dynamic change to explore the relationship of the above indicators of aGVHD and confirmed by in vitro experiments , TNF-α is capable of inducing HUVEC release EMPS . ① 21 routine allogeneic hematopoietic stem cell transplantation in patients with hematologic malignancies , was extracted from the peripheral blood of different points in time , the use of dynamic detection of peripheral blood flow cytometry CD62E EMPS level . ② using the enzyme-linked immunosorbent assay Ang-1, Ang-2, VEGF and TNF-α concentration . ( 3 ) using flow cytometry cultured human umbilical vein endothelial cells (HUVEC) after induction of TNF-α ( different concentrations and different time ) , the level of CD62 EMPS release . Results ① flow cytometry shows aGVHD group and no aGVHD group the EMPS water each time point average higher than the normal control group ( P lt; 0.05 ) 21d EMPS content than no of aGVHD group the aGVHD group was significantly higher (P lt ; 0.05 ) , while the the EMPS level of aGVHD than aGVHD group 21d significantly decreased ( P lt; 0.05 ) , no significant differences in other point in time . ② The ELISA shows , no of aGVHD group of Ang - 1 in 7d than aGVHD group increased ( P lt ; 0.05 ) , of aGVHD in 7d , 28d when EMPS level appear upward trend , but in aGVHD occurs significantly with decreased ( P lt ; 0.05 ) ; of aGVHD group of Ang- 2 in the 21d elevated compared with no aGVHD group (P lt; 0.05); aGVHD group , Ang-2 levels in the 21d with aGVHD was significantly higher ( P lt ; 0.01) ; the aGVHD group of VEGF concentration in 7d significantly decreased , after gradually returning to normal . ③ with TNF-α (100ng/ml, 36h) significantly induced HUVEC produce EMPS . Conclusion : ① EMPS and Ang - 2 in HSCT after surgery was significantly higher than the pre-transplant the aGVHD group with no aGVHD group significant differences , suggesting that the degree of endothelial injury in patients after allo-HSCT may be associated with the occurrence of aGVHD closely related . ② Ang-1.TNF-α and VEGF may reflect endothelial cell injury after transplantation , pending further study of its relationship with aGVHD .
