Irritable bowel syndrome(IBS)is a chronic disease characterized by abnormality of the dynamics and feeling of the gastrointestinal tract. It is characterized by long-term or recurrent abdominal pain and abdominal distention along with altered bowel habits and evacuation in clinical. It is a functional disease of bowel and nowadays is lack of the abnormal index in morphology and in bacteriology and biochemistry. The clinical manifestations of IBS are diverse and complex which are often dominated by colon symptoms as well as digestive tract and body.Irritable bowel syndrome(IBS)is distributed extensively. Although the incidence of IBS is up to 10%-20% in the word, its underlying mechanisms remain unclear. Maybe it correlates with a lot of factors, such as visceral hypersensitivity, alterations in bowel movements, the disordered function of movement out of stomach intestine, the abnormal function of automatic nervous system, diet, infection, psychology (long-range or excessive anxiety, depression, nervous) and so on.Some studies suggest that one of the reasons of IBS may be concerned with visceral hypersensitivity. Numerous relevant researches about IBS always involved in dorsal root ganglion, especially in dorsal root ganglion electrophysiology. But dorsal root ganglion is mainly concerned with the body sensations. IBS is internal organs - gastrointestinal function disorder, and of visceral sensation allergic related diseases. Visceral afferent neuron is almost located in nodose ganglion, which can innervate the intestinal tract, kidney, stomach and lung. In this study, using neural tracer method we explored the afferent difference of visceral sensation between IBS rats and normal rats, which provide basic theory of experiment for IBS and other intestinal disease.We made animal model of IBS by inducing inflammation with chemical reagent-TNBS, and investigated the projection of the ascending colon sensory afferent nerve to NG and DRG (L1-L6) by using HRP reverse tracing.The statistics result (SPSS 11.5) of the number of TMB staining masculine cells is followed:1 The number of the TMB positive cells of the group of IBS model had no difference with that of the normal group when HRP were injected in ascending colon (p > 0.05);The disposition of TMB positive labeled cell is diffuse, without fixed area. The long axis of the cells was about 10~30μM.2 The number of TMB positive labeled cell has no difference (p > 0.05) in the inner block from L1 to L6 on DRG. between the group of IBS model and the normal group.The peak of the number of the staining masculine cells was most localized at L4, L5 and L6 between the groups of the normal rat and the IBS model with HRP injected in ascending colon, their disposition is diffuse, without fixed area. The long axis of the cells was about 10~30μM.
