The Influence of p55PIK on Tumors Angiogenesis

Objective: To study p55PIK on tumor cells of vascular endothelial growth factor (VEGF) secretion, and its tumor cells into nude mice to investigate the relationship between tumor p55PIK with angiogenesis . Methods: adenovirus infected tumor cells to carry coded p55PIK gene differences; RealTime-PCR ( fluorescent quantitative PCR) to detect the level of VEGF mRNA in the tumor cells of the adenovirus infection empty adenovirus infection p55PIK expression by Western blot method of to detect differential expression of VEGF protein levels in tumor cells ; and planting in nude mice with colon cancer cells (HT-29) infection the empty adenovirus infection p55PIK adenovirus , of observation p55PIK tumorigenicity ; xenografts do the immunohistochemistry check, compare p55PIK VEGF and CD31 expression levels in the organization . Results : intracellular overexpression p55PIK VEGF in tumor cells mRNA and protein expression levels significantly increased; significantly enhanced tumorigenicity , faster tumor growth rate , size , weight than the group of empty adenovirus infection , overexpression VEGF and CD31 expression levels were significantly higher in the tumor tissue of the p55PIK group . Conclusion : p55PIK highly expressed on tumor cells of VEGF generated influential , high expression of the tumor cells in p55PIK tumorigenicity enhancement may be by increasing the level of VEGF to promote tumor angiogenesis to achieve .
