The Investigation about the Correlation of DcR3 with Lymphangiogenesis and Lymphatic Metastasis in Breast Cancer

Objective To study the decoy receptor 3 (DcR3) the relationship between lymphangiogenesis and investigate DcR3 role in breast cancer lymph node metastasis. Methods Immunohistochemical SP method detected 41 cases of breast carcinoma and adjacent normal tissue, DcR3 protein expression in the lymph nodes, lymphatic and D2-40 labeling of these organizations to observe the lymphatic microvessel characteristics calculated micro lymphatic density (MLVD). Using real-time quantitative PCR method for the detection of breast cancer tissue and adjacent normal tissue DcR3 mRNA expression levels, and to analyze the relationship between DcR3 and lymphatic microvessel density. Results Breast DcR3 expression was 58.5% in adjacent normal breast tissue DcR3 positive rate was 24.4%. DcR3-positive rate of lymph node metastasis (14/21) 66.6%, metastasis-free lymph node group DcR3-positive rate (7/20) 40%. Breast Cancer DcR3 expression was significantly higher than in adjacent normal breast tissue (P lt; 0.05), DcR3 expression is associated with lymph node metastasis (P lt; 0.05). Breast Cancer MLVD mean (9.266 ± 6.470) compared with normal breast tissue MLVD mean (2.773 ± 1.571) was significantly higher (P lt; 0.001); tumor surrounding MLVD mean (9.978 ± 5.743), the tumor center MLVD mean (8.554 ± 5.124) compared, the difference was not statistically significance (P gt; 0.05); breast cancer metastasis to lymph the organization MLVD mean (9.855 ± 5.542) than non-metastatic lymph node tissues MLVD mean (1.810 ± 1.611) was significantly higher (P lt; 0.000). Breast Cancer MLVD expression values ??and corresponding DcR3 protein staining results showed a significant positive correlation (r = 0.717, P = 0.000). DcR3 mRNA expression in breast cancer tissue (3.988 ± 3.706) higher compared with the corresponding normal breast tissue expression levels (2.291 ± 2.717) (P = 0.001). DcR3 mRNA in lymph node metastasis group expression levels (5.061 ± 2.970) than those without metastasis lymph DcR3 mRNA expression (2.042 ± 1.930) increased (P = 0.002). DcR3 mRNA expression in breast cancer tissue values ??corresponding DcR3 protein was significantly positively correlated (r = 0.629, P = 0.000). DcR3 mRNA in breast cancer tissue MLVD results showed a significant positive correlation (r = 0.698, P = 0.003). Conclusion: DcR3 higher expression rate in breast cancer, especially in cancer tissues with lymph node metastasis was significantly higher. DcR3 expression of D2-40 marker in breast cancer tissue micro lymphatic density into a positive correlation, suggesting DcR3 expression and lymphangiogenesis positively correlated, DcR3 to promote breast cancer lymph node metastasis may be through the induced lymphangiogenesis generation increases lymphatic metastasis of opportunities.
