Study of the Bloodcell Tissue Factor Expression of the Patients with Arterious or Venous Thrombus in Limbs

Objective: To investigate the blood cell-derived tissue factor gene and protein levels of expression and limb arterial and venous thrombosis. Methods: A case-control study, collected Fujian Provincial Hospital 60 cases of limb thrombosis confirmed by angiography as the case group, which limb arterial thrombosis 30 patients with deep venous thrombosis in patients with 30 cases over the same period in our hospital examination normal 30 cases as the control group; statistics of all study age (AGE), gender (SEX), fasting plasma glucose (FPG), triglyceride (TG), total cholesterol (TC), white blood cell count (WBC), neutrophil percentage ( N%), platelet count (PLT), total protein (TP), albumin (ALB), globulin (GLB), alanine aminotransferase (ALT), aspartate aminotransferase (AST), high-density lipoprotein ( HDL), low density lipoprotein (LDL), blood urea nitrogen (BUN), creatinine (Scr) and other clinical indicators. Using semi-quantitative reverse transcription - polymerase chain reaction (RT-PCR) method group of arterial thrombosis, venous thrombosis patients and control group members peripheral blood cells of tissue factor detection, UV gel imaging system scan imaging and semi-quantitative analysis, understanding of the target gene expression; using enzyme-linked immunosorbent assay (ELISA) detection study plasma levels of TF; using SPSS13.0 software to analyze data. Results: 1. Cases and control groups in age, gender, FPG, TG, TC, WBC, N%, PLT, TP, ALB, GLB, ALT,, AST, and HDL, LDL, BUN, Scr differences were not statistically significance, (P gt; 0.05). 2 with the control group (0.24 ± 0.09) compared to arterial thrombosis group (0.43 ± 0.10), the blood cells of the venous thrombosis group (0.39 ± 0.11) TF mRNA expression levels were significantly increased, (P lt; 0.05), the difference was statistically significance; arterial thrombosis group (0.43 ± 0.10) and venous thrombosis group (0.39 ± 0.11) compared to the P value of 0.166, the difference was not statistically significant. 3. Compared with the control group, arterial thrombosis group with venous thrombosis group, the plasma levels of TF was significantly higher (P lt; 0.05), the difference was statistically significant; Compared vein thrombosis, arterial thrombosis plasma TF levels, but The difference was not statistically significant, (P gt; 0.05). 4.Pearson correlation analysis showed: the group of arterial thrombosis, venous thrombosis patients blood cells TF mRNA expression levels in the plasma levels exist positive correlation, correlation coefficients were 0.890,0.937. Conclusions: 1. Within human blood cells express TF mRNA. 2. Limb arterial and venous thrombosis blood cells in patients with increased expression of tissue factor molecular level. 3. Limb arterial and venous thrombosis in patients with plasma tissue factor levels. Blood cells TFmRNA to express a positive correlation between plasma levels of TF. 5 high TF expression. Limbs moving the vein thrombosis occurrence of the disease with the source of blood cells.
