The Effects of Aerobic Exercise Training on Oxidative Stress and Ultrastructure of Pancreatic Organization in Diabetic Rats

Objective: This experiment which is based on the STZ-inducible diabetic rat model was to investigate the effects of aerobic exercise training on oxidative stress and ultrastructure of pancreatic organization in diabetic rats.To study the mechanism of the protective effects of aerobic exercise training on pancreatic mitochondria in diabetic rats.Methods: Thirty SD rats were randomly divided into three groups: normal control rats group, diabetic rats group and exercise diabetic group,with each group with 10 rats. Diabetic rats were estab1ished by abdominal injection of streptozotocin(STZ). Exercise groups were trained with 8 weeks swimming on a medium. After trained for eight weeks, the activities of superoxide dismutase(SOD), glutathion peroxidase(GPX), nitric oxide synthase(NOS)and the contents of malondialdehyde(MDA)and nitric oxide(NO)in the pancreatic mitochondria were observed respectively. The ultrastructure of pancreatic organization in rats was observed by electron microscope.Results: After trained for swimming for eight weeks, the plasma levels of fasting blood sugar(FPG)、triglycerides(TG)、cholesterol(TC) in diabetic rats group and exercise diabetic group were significantly higher than the normal control rats group(P﹤0.05); the weight of all the diabetic rats were significantly lower than normal control rats group, especially in the diabetic rats group(P﹤0.001). Exercise have a tendency to put on pounds, but it had no significant differences compared with diabetic rats group(P﹥0.05). The activities of SOD and GPx in pancreatic mitochondria in exercise diabetic group were increased more obviously than diabetic rats group (P<0.05),while the activity of NOS and the contents of MDA and NO were decreased obviously(P<0.05). The pancreas of the rats among all trial groups were observed by electron microscopy: in the normal control rats group, the Mitochondria of Pancreatic IsletβCells form neat,membrane integrity and cristae tidy; in the diabetic rats group, the Mitochondria of Pancreatic IsletβCells swell with vacuolelike changes, membrane dissolve, carina ruptured or disappearance and secretory granules reduce; by contrast, changes of the Mitochondria of Pancreatic IsletβCells of the exercise diabetic group rats, were less prominent than the ones of the diabetic rats group. The ultrastructure of pancreatic organization in exercise diabetic group was significantly reformed.Conclusions: Aerobic exercise training has protective effects on pancreas in diabetic rats, and can improve antioxidation ability to eliminate free radical, refine ultrastructure of pancreatic organization.
