Comparative Analysis on CTF and CEF Regimens as Postoperative Adjuvant Chemotherapy in the Treatment of Phase Ⅱ Breast Cancer

Objective:Breast cancer as a systemic disease,there are still recurrence and metastasis in some post-operative patients.Adjuvant chemotherapy as a systemic treatment plays an important role in the treatment of breast cancer.Anthracycline is one of the most effective drug treatment for breast cancer, the beginning of 1980,chemotherapy with doxorubicin is widely used in postoperative adjuvant chemotherapy for breast cancer.CAF regimen was standard programs for early breast cancer.Epirubicin and pirarubicin is a derivative of doxorubicin, its anti-tumor activity better than Adriamycin, and toxicity lighter. Currently the CTP with pirarubicin and the CEF with epirubicin is widely used in the treatment of breast cancer, and gradually replace the CAF regimens. This article is evaluated and compared the long-term efficacy and toxicity of the postoperative adjuvant chemotherapy of CTF and CEF in stage II breast cancer patients.Methods: Data of 83 women with stageII breast cancer treated in First Affiliated Hospital of Dalian Medical University between June 2004 and June 2006 were retrospectively analyzed. These patients were divided into two groups based on the regimen of the postoperative adjuvant chemotherapy:42 received CTF regimen(5-FU 500mg / m~2 iv d1 CTX 500mg / m~2 iv d1 THP50mg / m~2 iv d1) and 41 received CEF regimen(5-FU 500mg / m~2 iv d1 CTX 500mg / m~2 iv d1 E-ADM90mg / m~2 iv d1 ),with 3 weeks for each cycle , a total of 6 cycles. We used chi-square to analysis to analyze long-term efficacy and toxicity of two regimen for comparison.Results: 5-year survival were 100% in both CTF and CEF regimen, disease-free survival rate of CPF regimen is 92.9%, CEF regimen is 95.1%, the difference was not statistically significant, P > 0.05. Toxicity: The incidence of alopecia CEF regimen was significantly higher than the CTF regimen, 90%vs36%, P = 0.000; In the III / IV toxicity,there are all significant differences in both groups,leukopenia:CTF highter than CEF regimen ,6.7%vs 4.9% (P = 0.045); nausea and vomiting:CEF highter than CEF regimen, 12.2%vs2.4%,(P = 0.046); alopecia:CEF highter than CTF regimen, 24.4%vs0%,(P = 0.001).Conclusion: 1 There is no significant difference in the 5-year survival and disease-free survival on the CTF and the CEF regimen2 The incidence of alopecia CEF regimen was significantly higher than the CTF regimen3Ⅲ/Ⅳtoxicity: leukopenia in the CTF regimen highter than CEF regimen; nausea and vomiting、alopecia in the CEF regimen highter than CTF regimen
