Expression and Clinical Significance of PTEN and Caspase-7 in Gastric Cancer and Precancerous Lesion

Objective:Gastric cancer was one of the most common malignant tumor in China, It was the fourth in morbidity rate of malignant tumor and was the second in mortality rate of malignant tumor, early detection, diagnosis and treatment of Gastric cancer has been the hotspot and difficulty. By detecting the expression and correlation of PTEN, Caspase-7 protein in Gastric cancer and precancerous lesions to explore their potential participate pathogenesis about gastric cancer for early diagnosis of gastric cancer and provide a scientific basis for treatment.Methods:Immunohistochemistry SP was used to detect the expression of PTEN and Caspase-7 in 54 patients with gastric cancer tissue and 50 patients without gastric cancer tissue and analyzed the relation between PTEN and Caspase-7 with clinical pathological characteristics, Statistic analysis was completed with SPSS 11.5, all data were tested by x2-test or Spearman rank correlation test, p<0.05 was considered statistical signification.Results:1. The expression of PTEN in gastric cancer and without gastric carcinoma tissue Expression of PTEN protein located in the cytoplasm, yellow granules, The positive expression of PTEN protein in gastric cancer was 55.6% which significantly lower than 93.8% in without gastric cancer ,there were significant differences between gastric cancer and without gastric cancer (p<0.05).The positive expression of PTEN protein in without gastric cancer tissue including non-atrophic gastritis ,atrophic gastritis ,dysplasia was 100.0%, 93.8% and 78.6% which demonstrated the decreasing tendency. The positive expression of PTEN protein in dysplasia was significantly lower than in non-atrophic gastritis and atrophic gastritis. 2. The expression of Caspase-7 in gastric carcinoma and without gastric cancer tissueExpression of Caspase-7 protein mainly located in the cytoplasm, yellow granule.The positive expression of Caspase-7 protein in gastric cancer was 51.9% which significantly lower than 88.0% in without gastric cancer ,there were significant differences between gastric cancer and without gastric cancer (p<0.05). The positive expression of Caspase-7 protein in without gastric cancer tissue including non-atrophic gastritis, atrophic gastritis, dysplasia was95.0%,87.5%and78.6% which demonstrated the decreasing tendency .The positive expression of PTEN protein in non-atrophic gastritis、atrophic gastritis and dysplasia had little difference (p>0.05).3. The relationships between PTEN and different clinical characteristics of gastric cancerThe expression of PTEN in gastric cancer had no significant correlation with sex ,age, tumor size, tumor location (p>0.05), and had significant correlation with tumor differentiation, invasion depth, lymph node metastasis, clinical stage (p<0.05).4. The relationships between Caspase-7 and different clinical characteristics of gastric cancerThe expression of PTEN in gastric cancer had no significant correlation with sex, age, tumor size, tumor location, invasion depth, lymph node metastasis and clinical stage (P> 0.05),and had significant correlation with tumor differentiation (P <0.05).5. The relevance of PTEN and Caspase-7 in gastric cancerIn 30 cases which PTEN expressed positive in gastric cancer Caspase-7 expressed positive in 21 cases. In 24 cases which PTEN expressed Negative in gastric cancer Caspase-7 expressed negative in 7 cases, PTEN is associated with Caspase-7 (p﹤0.05).Conclusion:1. The low expression of PTEN may participate in the formation of gastric cancer and precancerous lesions. PTEN was correlated with tumor differentiation, invasion depth, lymph node metastasis, clinical stage.2. The low expression of Caspase-7 may participate in the formation of gastric cancer, Caspase-7 was correlated with tumor differentiation.3. The expression of PTEN and Caspase-7 was positively correlated in gastric cancer.
