Beta ig-h3 is a extra cellular matrix protein, which is secreted by many cells. It consists of four conserved internal homologous domains. It contains a Arg-Gly-Asp(RGD) sequence at C terminal that can combine integrin which is a glucoprotein locating at cell surface, then the complex mediates interaction between cell and extra cellular matrix, and mediates cell’adhesion, proliferation, migration, and differentiation. Beta ig-h3 gene is ubiquitously expressed in many human normal tissues except for brain tissue. It’s differently expressed in many tumor tissues. The RGD sequence can combine different intergins in different cells and tissues, then play different roles, it can combine interiginαvβ5,then promote colon cancer cell’migration; combines interiginα3β1,then promote hepatoma cells’invasion. The gene encoding beta ig-h3 protein locates in chromosome 5q31,that sometimes mutations or loses in many tumors and diseases. The mouse deleted of beta ig-h3 gene produced spontaneous tumor easily. Transfecting beta ig-h3 gene into Chinese Hamster Ovary (CHO) fibroblasts has been shown to markedly reduce their tumorigenicity in nude mice. That shows beta ig-h3 is associated with tumor. But expression of beta gi-h3 depends on the type of cell and tissue, also the effect of beta ig-h3 depends on the type of tumor, including promoting and suppression. Beta ig-h3 could promote adhersion and migration of cancer cell in colon cancer and hepatoma, and suppress neuroblastoma developing in vivo.Objective:We measured the expression of beta ig-h3 in gastric cancer tissue. Then supply helps for studying molecular mechanism of gastric tumorigenesis and the therapy target of gastric cancer.Method:Measured the expression of beta ig-h3 in gastric cancer tissue, paragastric carcinoma tissue and normal gastric tissue with immunohistochemistry. Every slice has been taken five pictures randomly, and measured the proportion of positive area in every picture with image-pro-plus software, then computed the average proportion of positive area of every patient and of every group. Evaluated statistic differences among the groups with statistic software SPSS 17.0.Result:The expression of beta ig-h3 is down-regulated in gastric cancer tissue, and up-regulated in normal gastric tissue and paragastric carcinoma tissue, the proportion of positive area of the three groups respectively is 0.1913±0.0814,0.1692±0.1287,0.0471. There is statistic difference among them(P<0.05). There is statistic difference between gastric carcinoma tissue and normal gastric tissue(P<0.05),there is statistic difference between gastric cancer tissue and paragastric carcinoma tissue(P<0.05). There is little statistic difference between normal gastric tissue and paragastric carcinoma tissue(P>0.05),and between high-grade adenocarcinoma and low-middle- grade adenocarcinoma(P>0.05),and among different types of gastric carcinoma.Conclusion:The expression of beta ig-h3 is down-regulated in gastric cancer tissue, there is no difference between high-grade adenocarcinoma and low-middle-grade adencorcinoma,beta ig-h3 maybe suppress the production and development of gastric cancer.
