Study on Expression of MMP-8 on Experimental Periapial Periodontitis is Infected by Enterococcus Faecalis

Objective: Study on expression of matrix metalloproteinase 8 in periapical periodontitis is infected by Enterococcus faecalis of rats model.Methods: Selected 21 adult male SD rats, the 18 were randomly divided into two experimental groups, A group of the rat right mandibular first molar occlusal opening marrow, the marrow directly exposed to the oral environment. We feeded them regularly during second weeks to sixth weeks. Then filmed their puple with X-ray and CBCT films at 2W、4W、6W , then separation of the first molar and its periapical tissue in right mandibular at second weeks, fouth weeks, sixth weeks. Then make some frozen sections and make an HE staining.Make an immunohistochemistry staining in order to observation of expression of matrix metalloproteinases-8.Use SPSS make an experiment result.Results:1.X-ray film and CBCTA group and B group the first molar apical area of right mandibular showed nothing changed and finded shadow at fourth weeks and 6weeks. Make sure that the periapical periodontitis of rats model by the pulpal sealed by Enterococcus faecalis is established . Group B,apical shadow is larger than Group A in the rat animal model of periapical lesions. Health group had nothing shadow .2. HE stainingA group and B group have a large number of inflammatory cell infiltration and formate abscess in right mandibular first molar and apical arear at 2 weeks. Two groups have a large number of lymphocytes at 4 weeks and 6 weeks. Health group had nothing changed. 3.Immunohistochemical stainingB group showed neutrophils and lymphocytes produced MMP-8 in periapical tissue at second weeks. B group have nothing positive cells at 4 weeks and 6 weeks. A group and health group have nothing positive cells at2、4、6 weeks.Conclusion: 1. The periapical periodontitis of rats model by the pulpal sealed by Enterococcus faecalis is established . 2. MMP-8 is expressed at 2 weeks group, suggesting that E. faecalis may enhance expression of MMP-8 in periapical tissue. Enterococcus faecalis may accelerate the process of development of periapical inflammation.
