Application of DNA Barcoding in Species Identification of Colocasieae (Araceae)

DNA barcoding the taxonomic research focus in recent years, it is a short sequence of DNA technology for identification of the species. The technology is still at an early stage in the study of plant taxa, slightly behind fauna. Some length 650bp mitochondrial cytochrome C oxidase subunit Ⅰ (COI) can effectively identify the many animal groups, the rate of addition to fungi, COI and mitochondrial gene evolution in plants is much slower than the rate of evolution in animals, is only applicable in some algae, but domestic and foreign experts to speed up the pace, looking for a plant kingdom versatility of DNA barcoding. In addition to species identification, DNA barcode technology can be found many new species and cryptic species, can significantly improve the accuracy and efficiency of field ecology study tour; the DNA barcode also can be applied to solve complex biological problems, for example, by correctly identified parasitic plant and determine the trophic relationships. The taro family of plants is an important group of Araceae, but some taxonomic status, system location, there is a big controversy. With the means of molecular biology, including DNA barcode technology, combined with morphological classification method to clarify the phylogenetic relationships of the taro family of plants and to correctly identify the species, taro family plant genetics and breeding, germplasm conservation and sustainable use has important significance. This study to the the taro family of 4 genera and 14 species of 78 samples for the study material, as a candidate gene fragments using matK, rbcL, trnH-psbA, ITS, through genetic distance, sequence homology analysis, GC% content, the system tree and many other kinds of analytical methods to distinguish and identification of species, and discussion of individual fragments and combinations of fragments as the taro family of plant DNA barcode adaptability, the Plantae DNA barcode research provide new information. The result: better sequencing results; addition to ITS, this study suggests that the combination of the two sequence fragments rbcL trnH-psbA plant DNA barcode as taro family, distinction and identification of plant taro family. DNA barcoding and morphological classification data analysis showed: Ono taro can separate case; the heterochromatic the taro and Yunnan taro genetic relationship very far support the the Leis the taro and Yunnan taro as separate species, should not be a synonym for fake taro; The support both Gong's the taro and Lee Taro as a separate species, should not become the name of the southern Yunnan taro exclusive; support purple rod taro as Lee Taro variants.
