Objective:From the respects of pharmaceutical and pharmacodynamic to study the compatibility mechanism of Copitis-Evodia .Methods: From pharmaceutical respect, using the method of HPLC, with the contents of Jatrorrhizine hydrochloride, Hydrochloric acid palmatine, Berberine, Evodiamine and Rutaecarpine as index components, determinated of the content of the sample in different proportions, establishing quality standards. Using the method of HPLC, according to HPLC profiles given parameters, establishing Copitis-Evodia HPLC spectrum. From pharmacodynamic respect, with the indicators of gastric ulcer, gastric acid secretion , tolerance and diarrhea. Studing tissue distribution in mice, to explore the pharmacological effect laying the material basis.Results: 1.Using the method of HPLC, establishing quality standards and HPLC spectrum of four Copitis-Evodia compatibility groups. The Jatrorrhizine hydrochloride, Hydrochloric acid palmatine, Berberine, Evodiamine and Rutaecarpine contents of four groups (6:1,2:1,1:1,1:6) were not less than 14.51 mg·g-1、32.57 mg·g-1、108.8 mg·g-1、1.435 mg·g-1、1.669 mg·g-1;14.50 mg·g-1、37.49 mg·g-1、130.7 mg·g-1、3.271 mg·g-1、3.307 mg·g-1;12.88 mg·g-1、21.79 mg·g-1、71.05 mg·g-1、3.608 mg·g-1、3.048 mg·g-1;2.618 mg·g-1、6.476 mg·g-1、23.69 mg·g-1、5.246 mg·g-1、5.576 mg·g-1.By 4 ratio the test parpmeters derived from HPLC profiles, calibrated the 16 peaks, with Berberine as an index component, to determine its stability of quality with peak height ratio.2. Through efficacy trials , in terms of anti-ulcer, 6:1 group was best,2:1 was second; four groups were not significant effect of gastric acid inhibition;2:1 group had the best heat tolerance;1:1 group had the best antidiarrheal effect,2:1 group was second. In addition , 1:6 group had more significantly analgesic effect.3.By the test of tissue distribution, we can detectied Hydrochloric acid palmatine and Berberine in heart, liver, spleen, lung, kidney, stomach, small intestine, and large intestine, gastrointestinal system had higher organization concentration,when 1 hour, organization concentration was the highest.Conclutions: 1.Using the method of HPLC, establishing quality standards and HPLC spectrum of four Copitis-Evodia compatibility groups. The Jatrorrhizine hydrochloride, Hydrochloric acid palmatine, Berberine, Evodiamine and Rutaecarpine contents of four groups (6:1,2:1,1:1,1:6) were not less than 14.51 mg·g-1、32.57 mg·g-1、108.8 mg·g-1、1.435 mg·g-1、1.669 mg·g-1;14.50 mg·g-1、37.49 mg·g-1、130.7 mg·g-1、3.271 mg·g-1、3.307 mg·g-1;12.88 mg·g-1、21.79 mg·g-1、71.05 mg·g-1、3.608 mg·g-1、3.048 mg·g-1;2.618 mg·g-1、6.476 mg·g-1、23.69 mg·g-1、5.246 mg·g-1、5.576 mg·g-1.By 4 ratio the test parpmeters derived from HPLC profiles, calibrated the 16 peaks, with Berberine as an index component, the method had good stability and repeatability, as a qualitative basis.2. Through efficacy trials , the effects were different, 6:1 group had the best anti-ulcer effect, 2:1 group had the best heat tolerance and more significantly analgesic effect, 1:1 group had the best antidiarrheal effect. In addition , 1:6 group had more significantly analgesic effect. 3.By the test of tissue distribution, we can detectied Hydrochloric acid palmatine and Berberine in heart, liver, spleen, lung, kidney, stomach, small intestine, and large intestine, gastrointestinal system had higher organization concentration,when 1 hour, organization concentration was the highest, that showed Palmatine and Berberine were widely distributed and metabolism in mice.
