Correlation Analysis and QTL Mapping for Quality Traits in Wheat

To offer theoretical reference for wheat quality genetic marker assisted select breeding and fine mapping of related genes, in the present study, a total of 209 doubled-haploid lines (DH) derived from the cross Huapei 3/Luomai 4, which were planted in Beijing and Zhengzhou locations in 2008, were used to analyse correlation among quality parameters and to identify QTLs for flour quality parameters. Besides, mixograph parameters were analysed by using three-year datas (2006-2008). Quality traits include 13 quality parameters:swelling power, swelling index of glutenin, solvent retention capacity, mixograph parameters. Composite interval mapping was used in QTL mapping. The results are described below:(1) The SIG results is negatively highly significantly related to the peak value. Significant positive correlation was observed among four SRCs. The mixing stabitity time,7 minute width and 7 minute peak were positively highly significantly correlated with the mixing time, the peak value and weaking slope were negativey highly significantly related to the mixing time. The 7 minute width and 7 minute peak were positively highly significantly or positively significantly correlated with the mixing stabitity time, the peak value and weaking slope were negativey highly significantly related to the mixing stabitity time. The peak value is positively highly significantly related to the peak width. The peak value and the peak width were positively highly significantly correlated to the 7 minute peak. The 7 minute width was positively highly significantly correlated with the 7 minute peak.(2) Nine QTLs were detected for swelling power, swelling index of glutenin, solvent retention capacity on chromosomes IB,3B,4A,4B and 7A, no common QTLs was detected in two areas. Three QTLs for swelling power were located in different intervals on chromosome 4A, explaining 37.92% of the phenotypic variance. Two QTLs for SIG were detected on chromosomes IB and 7A, which explaining 13.47% of the phenotypic variance. Two QTLs for the water SRC were detected in Beijing, which explaining 5.16% and 5.99% of phenotypic variance, respectively. Two QTLs for the lactic acid SRC were detected in Beijing, accounting for 8.6% and 11.02% of phenotypic variance, respectively. One QTL for sucrose SRC was detected in Henan and explained 6.15% of the phenotypic variance.(3) Forty nine QTLs were mapped for Mixograph parameters on chromosomes 1A, IB, IDS,2A,2B,3AL,3B,4A,4B,5AS,5B,5DL,6A, 6B,7A,7B and 7D. Seven QTLs were stable and were located on chromosomes 1A, 1B and 3AL. The marker interval Barc287~Gwml6 on chromosomes 1A influenced peak value, accounting for 5.31%~8.78% of phenotypic variance. The marker interval Wmc2-Wmc527 on chromosomes 3AL influenced peak value, explaining 4.37%~5.27% of phenotypic variance. The marker interval Xcau469-Barc187, Barc187~Barc302, Barc181~Barc119, Barc312~Barc120 and Barc120~Ksm145 were on chromosomes 1B. Xcau469-Barc187 influenced peak value, mixing time, mixing stabitity time,7 minute width and 7 minute peak, accounting for 5.79%~48.47% of phenotypic variance. Barc187~Barc302 influenced peak value, mixing time, peak width and 7 minute width, explaining 5.64% 27.15% of phenotypic variance. Barc181-Barc119 influenced mixing stabitity time,7 minute width and 7 minute peak and weaking slope, accounting for 7.09%~22.23% of phenotypic variance. Barc312-Barc120 influenced peak width, explaining 11.01%~17.55% of phenotypic variance. Barc 120~Ksm 145 influenced mixing stabitity time, accounting for 7.3% 15.03% of phenotypic variance.
