The Preliminary Study on Antitumor Mechanism of Aqueous Extract of Taxus Chinensis in Vitro

Objective:To research the extensive anti-tumor effect of T.chinensis var in vitro, to identify its difference of anti-tumor mechanism with paclitaxel, and to explore its possible anti-tumor mechanism.Methods:Extracted from T.chinensis var by cooked with water:the cell proliferation effect of T.chinensis var on vitro cultured human lung adenocarcinoma A549 cell、gastric cancer SGC-7901 cell and breast cancer MCF-7 cell are detected by MTT method; Time-effect relationship on inhibition of these cells is observed by growth curve; effects on Induced these cells’apoptosis are detected by flow cytometry; morphology of these cells are observed by ordinary optical microscope; expression of apoptosis-related protein P38 and P-JNK are detected by Western blot.Result:(1) The yield rate of the aqueous extract of T.chinensis var is 8.25%. (2) MTT method finds that the aqueous extract of T.chinensis var has inhibitory effect on proliferation of A549、SCG-7901 and MCF-7, and the OD values of the dose groups (500μg/ml、600μg/ml、700μg/ml、800μg/ml、900μg/ml、1000μg/ml) compared with the control group, shows significantly statistical difference (P<0.05 or P<0.01). besides, there is no significant difference in half inhibited rates (IC50), which means different tumor cells have the same sensitivity to it. And paclitaxel also has inhibitory effect on proliferation of these cells, and the OD values in the dose groups (1.0mg/ml、1.5mg/ml、2.0mg/ml、2.5mg/ml、3.0mg/ml、3.5mg/ml) compared with the control group, shows significantly statistical difference too(shows significant difference too)(P<0.05 or P<0.01). However, it shows different half inhibited rates (IC50) (P<0.05), which means different tumor cells have different sensitivity to it. (3) Growth curve finds that there is a certain time-effect relationship on the inhibitory of these cells’proliferation after treated by the aqueous extract of T.chinensis var. (4) Optical microscope finds that tumor cells treated by the aqueous extract of T.chinensis var shows the typical manifestations of apoptosis; however, those treated by Taxol, mainly shows the expression of necrosis. (5) Western blot detection finds that the expression of P38 protein is unchanged, but P-JNK protein is decreasing with the increase of drug concentration in cell A549 after treated by the aqueous extract of T.chinensis var.Conclusions:(1) The aqueous extract of T.chinensis var has inhibitory effect on proliferation of tumor cell, and with a certain dose-effect relationship and time-effect relationship. (2) The aqueous extract of T.chinensis var has different anti-tumor mechanism with paclitaxel, and its mechanism is possibly related to inducing the apoptosis of tumor cell.
