Ojective:To investigate the protective effects of DIDS on apoptosis of cultured hippocampal neurons induced by SIN-1 (a donor of NO) and the effect of DIDS on expression of PARP-1 and AIRMethods:The primary cultured neurons of 12days old from SD rats wre randomly divided into three groups:control groups、SIN grops and SIN-1+DIDS grops.The duration of both SIN-1 and DIDS effect on neurons for 18h.The neuronal viability was tested by MTT assay.Fluorescent staining with Hoechst33258 was used to analyze apoptosis.The methods of Western blot and immunocytochemical fluorescent stain were used to detect the expression of PARP-1 and AIF.Results:(1)SIN-1 may induced cultured hippocampal neuronal apoptosis with dose-dependence and 1.0mmol/L of SIN-1 can induce neuronal apoptosis obviously. (2)The chloride channel blockers of DIDS could inhance the neuronal viabilities and inhibitted the neuronal apoptosis induced by SIN-1.(3)Protein PARP-1 and AIF increased expression after the SIN-1 induce neuronal apoptosis.(4)Chloride channel blockers DIDS could significantly reduce the expression of PARP-1 and AIF induced by SIN-1.Conclusion:Chloride channel blocker DIDS perheps can save injured neuron by the way of influence PARP-1/AIF in which of non-caspase dependent aopotosis pathway.
