Expression and Correlation of Caspase-3 and Cx43 in Myocardial Infarction with Different Narrow Degree Coronary Artery

Objective: To detect the expression levels of Caspase-3 and Cx43 and the correlation between them in myocardial infarction with different narrow degree coronary artery.To research Change rule and diagnostic significance of Caspase-3 and Cx43 in Coronary heart disease cases with myocardial infarction.Method: Twenty-five specimens of human myocardium of left ventricular were obtained from autopsy patients in dalian from 2008 to 2009.The patients in this study were aged from 39 to 84,average 59.64.Those specimens were diagnosed different levels of coronary atherosclerosis in left anterior descending and levorotatory and myocardial infarction. According to the different coronary artery stenosis degree which is divided into four groups: namely group A (≤25%) 4 samples, group B (25% -50%) 6 samples, group C (50% -75%) 7 samples and D groups (≥75%) 8 samples.All speciments were fixed in 10 % paraformaldehyde,dehydrated,waxed,embed in paraffin.The expression of Caspase-3 and Cx43 were examined by immunohistochemical technique.Result: 1. Immunohistochemistry of Caspase-3:Caspase-3 protein can be tested positive expression with tan or brown particle in myocardium cytoplasm of each group. There was an obvious difference of Caspase-3 espression level in A,B,C and D group(P < 0.05).But there was no significant difference of Caspase-3 espression level between C and D group(P>0.05).2. Immunohistochemistry of Cx43:The positive expression of Cx43 have obvious change in each group. Cx43 was not found in the necrotic zone.The Cx43 was totally disappeared in border zone and only a few tan or brown particle were seen in longitudinally oriented side of the cardiac muscle cells or show pointlike distributed in cytoplasm. There was an obvious difference of Cx43 expression level between A,B,C group and B,C,D group(P < 0.05).But there was no significant difference of Cx43 espression level between A,B and C,D group(P>0.05).3. Correlation comparison of Caspase-3 and cx43: With narrow degree of aggravating in all samples,Caspase - 3 positive express cells number is on the increase while Cx43 positive expressed cells number is in gradually diminishing.But there is not obvious negative correlation between Caspase-3 and Cx43 expression in myocardial infarction specimens(rs=-0.148 ,P=0.479).Conclusion:1.The expression of Caspase-3 and Cx43 were observed and abnormal distribution in early myocardial infarction. They can be used as a diagnosis of index in the early myocardial infarction.2. In the myocardial infarction cases with the narrow degrees of coronary artery stenosis increased,the expression level of Caspase–3 and Cx43 has significant difference (P < 0.05).
