The Expression and Relationships of Bmi-1 and c-myc in Diffuse Large B-cell Lymphoma Tissues

【objective】Malignant lymphoma is from lymph nodes and lymph tissue.The etiological factors and pathological mechanisms are still ambiguous,they could be related with the immunologic disfunction and the disequilibrium of cell proliferation and apoptosis. Bmi-1 gene is the first identification of the PcG gene family,and is also one of the functional PcG who is make a important role in maintain neural stem cells, hematopoietic stem cells, cancer stem cells proliferation.A number of studies show that Bmi-1 gene for the maintenance of hematopoietic stem cell self-renewal and differentiation potential to play an important role, and in a variety of tumors with high expression. c-myc oncogene is a nuclear protein transcription factors ,it play a very important role in cell cycle and the process of malignant transformation of cells and high expression can lead to apoptosis obstacles. So, we observed the expression of bmi-1 and c-myc in 60 diffuse large B-cell lymphoma tissues and analyzed the correlation between them, in order to further explore the pathogenesis of malignant lymphoma and provide a theoretical basis for prognosis.【Methed】1.The expressions of Bmi-1 and c-myc were detected in 60 diffuse large B-cell lymphoma wax samples by immunohistochemical method respectively. 2.The expressions of Bmi-1 and c-myc were detected in 20 reactive hyperplasia lymphoid tissues wax samples by immunohistochemical method respectively. 3.The relationships of Bmi-1 and c-myc and their correlations to clinical characterizes were analyzed in diffuse large B-cell lymphoma.【result】1.The positive rate of Bmi-1 protein was 65.0% in DLBCL,which was 10% in reactive hyperplasia lymphoid tissues .There were significant differences between them(P <0.01). The positive rate of c-myc protein was75.0% in DLBCL, which was 25% in reactive hyperplasia lymphoid tissues .There were significant differences between them(P <0.01). 2.The expression of Bmi-1 was positively correlated to that of c-myc in DLBCL,the expression of Bmi-1and c-myc were related with the clinical stage and IPI score(P <0.01),but there was no obvious correlation between sex,age,extranodal involvement and B symptoms(P> 0.05)【conclusion】Bmi-1 and c-myc are found to be highly expressed in the tissues of DLBCL,they may play a role in the occurrence and development of DLBCL. Bmi-1 and c-myc could be used as prognostic markers.
