The Influence of Bile Duct Partial Obstruction on Guinea Pigs’ Sphincter of oddi in Early Stage

Objective:To investigate the changes of movement function, morphology and electrophysiology of sphincter of oddi of bile duct pure partial obstruction In early stage.Methods:Twenty-four England short-hair guinea pigs were randomly divided into three groups. Control group (Group A, n=8) was underwent celiotomy and purely palpated the common bile duct (CBD).Experimental group(Group B,n=8)was the partial obstruction of CBD by ligaturing both a cannula(e.d0.05mm) and the common bile duct near to its bifurcation following drawn the cannula. A.Drugs group(GroupC,n=8) was injected the for salviae miltiorrhiza(ISM)(0.1ml/d) for 7 days after the partial obstruction of CBD as group B. After feeding one week myoelectronic activity of the sphincter of oddi(MASO), pressure of the sphincter of oddi (SO) and bile duct (BDP)were measured respectively by subserosal contacted electrode and 4/25 power transforming system.The pathology changes,and TGF-β1 and SR of SO were observed.Results:In the group B and group C,the TBIL,DBIL,GGT,ALT and the CBD diameter and the basal pressure of CBD were higher than that in the group A. In the group B and group C the basal pressure of SO were significantly increased than that in the group A., whereas the voltage amplitude of slow waves and spikes in the groupB and group C decreased significantly than that in the group A. However there were no obvious change for the frequency of SO contraction among 3 group. Compared with the control group, the transforming growth factor--β1 (TGF--β1)and the pathology grade of SO was significantly increased in the group B and group C but the secretin receptor(SR) was invariant. However, the SO basal pressure, the voltage amplitude of slow waves and spike of SO in the group C were significantly improved than that in group B.Conclusion:The partial obstuction of CBD caused significantly changes of SO dynamic、myoelectric activities and TGF-β1.It was suggested that the changes may be the important factors to cause acute bile pancreatitis.The injection salviae miltiorrhiza(ISM) can improve the Sphincter of Oddi dysfunction(SOD).
