Campylobacter jejuni (C.jejuni) is the most frequent cause of bacterial diarrhea.The important thing is that.Recent research show that most of the development of Guillain-Barrésyndrome (GBS) have relations with the preceding infection of the certain C. jejuni strains.With development of neurophysiological and pathological studies ,it has led to a reclassification of GBS into acute inflammatory demyelinating polyneuropathy(AIDP)、acute motor axonal neuropathy(AMAN)、acute motor and sensory axonal neuropathy(AMSAN)、Miller-Fisher syndrome (MFS). The development of GBS are associated with certain C.jejuni strains with specific serotype.In our study, we analyze mutation of 3 GBS-associated C.jejuni strains which isolated from GBS patients in our northern area of China. We search the different gene flagments between NCTC11168 and GBS-associated C.jejuni such as cj1119-cj1152, cj1293-cj1342 and cj1413c-cj1448c to find out the relevance between the sequence character and ability to cause GBS as also as the markers for local GBS associated C.jejuni.MethodsThe C,jejuni strains used were isolated from stools of AMAN patients from the northern area of China.The serotypes are respectively Penner O:19, Penner O:2, Penner O:5.The detected complete genome sequences of the serotypes of Penner O:2, Penner O:5, Penner O:19.Use the Sequence Alignment software to contrast the complete genome sequences of Penner O:2, Penner O:5, Penner O:19 with the complete genome sequences of NCTC11168 from Genbank to find the mutant bases.Look over the supposed and definite mutational gene fragments in the literatures,find the mutant bases in these gene fragments in the complete genome sequences contrast of the 3 C.jejuni strains of pennerO:2,pennerO:5 ,pennerO:19 and NCTC11168.After finding the mutant bases of the 3 C.jejuni strains,find out the mutation of the 3 C.jejuni strains in the same basic site.It divides into three circs:⑴all of the 3 C.jejuni strains have the mutation in the same basic site;⑵two discretional strains of the 3 C.jejuni strains have the mutation in the same basic site;⑶only one strain of the 3 C.jejuni strains have the mutation in the same basic site.According the count of the mutant bases in the gene fragments to arrange ordering of the mutant gene fragments.According the ordering and literatures to divide the mutant gene fragments into five sorts:⑴the definite mutant gene fragments by experimentation in the literatures;⑵the most significant mutant gene fragments according the ordering;⑶the significant mutant gene fragments according the ordering;⑷the little significant mutant gene fragments according the ordering;⑸no mutant gene fragments according the orderingAnalysis the differences in the 3 strsins and the significance of gene mutation.Result1. Campare with NCTC 11168 gene sequence,lulei strain has 13183 mutant bases, qiaoyuntao strain has 3624 mutant bases, zhanxing strain has 16505 mutant bases.2. Find 137 supposed and definite mutant gene fragments caused GBS from the literatures.There are 22 mutant gene fragments which have the same nucleotide mutation in the same basic site of all of lulei strain,qiaoyuntao strain and zhanxing strain . There are 50 mutant gene fragments which have the same nucleotide mutation in the same basic site of two discretional strains of the 3 C.jejuni strains. There are 40 mutant gene fragments which don’t have the same nucleotide mutation in the same basic site of the 3 C.jejuni strains. There are 25 gene fragments which don’t have the nucleotide mutation in all of the the 3 C.jejuni strains.3. The definite mutant genes by experiment caused GBS are galE、neuB1、cst-Ⅱand waaF in the literatures.According the ordering to suppose the most significant mutant genes have 20, the significant mutant genes have 50, the little significant mutant genes have 40, the insignificant mutant genes have 23.ConclusionFind 137 supposed and definite mutant gene fragments caused GBS from the literatures. The definite mutant genes by experiment caused GBS are galE、neuB1、cst-Ⅱand waaF in the literatures.According the ordering to suppose the most significant mutant genes have 20, the significant mutant genes have 50, the little significant mutant genes have 40, the insignificant mutant genes have 23.But there is no mutation of neuB1、cst-II and so on in lulei strain,qiaoyuntao strain and zhanxing strain.One reason is maybe no mutation of neuB1、cst-II and so on in local C.jejuni strains caused GBS,supposed the character of local C.jejuni strains,it need more local C.jejuni strains to validate.The other reason is that GBS is maybe not single gene determined,maybe the resule of common effect of some genes.The same mutation in the local C.jejuni strains can make significant change of the coding protein,change the function of those proteins,become molecular biology basis of high pathogenicity in local C.jejuni strains and high GBS incidence rate in the northern area of China.Our study established the foundation for exploring the biologically characteristic of local GBS-associated C.jejuni stains,surveying the GBS-associated C.jejuni stains,preparation of the vaccine.
