The Significance of Expression and Genetic Mutation of Cx26, Cx32 in the Pathological Skin Scar and Scar Carcinoma

Objective:To explore the significance of expression and genetic mutation of Cx26,Cx32 in the pathological skin scar and scar carcinoma tissues. Methods:We used pathological skin scar tissues and skin scar carcinoma tissues to compare with normal skin tissues. The expression of Cx26 and Cx32 proteins were detected by the immunohistochemical method of S-P. And the expression of Cx26mRNA and Cx32mRNA were detected by in situ hybridization. The expression (optical density and positive area) of Cx26,Cx32 proteins and Cx26mRNA,Cx32mRNA were detected respectively by computerized image-analysis in normal skin epidermis, pathological skin scar epithelium and scar carcinoma tissues. All data was input into the computer and statistically analyzed with SPSS(16.0) software. On the other hand, analyze the frequencies and characteristics of Cx26 and Cx32gene mutations. DNA was extracted from Chinese patients with the choosen 28 samples fixed in10% formalin and Paraffin embedded(14 pathological skin scar and 14 pathological skin scar carcinoma tissues)and screened for mutations in Cx26 and Cx32 genes by direct sequencing. Results:(1) Cx26 and its mRNA, Cx32 and its mRNA showed weakly positive or negative expression on the epithelium of scar carcinoma tissues. Compared with normal skin group and skin scar group, the expression (positive area and optical density) of Cx26 and its mRNA, Cx32 and its mRNA in scar carcinoma group were statistically significant (P<0.01). (2) Cx26 and its mRNA showed strong positive expression in pathological skin scar epitheliums, Compared with normal skin group, the expression (positive area and optical density) of Cx26 and its mRNA in scar group were statistically significant (P<0.01). (3) Cx32mRNA showed positive expression in pathological skin scar epitheliums, Compared with skin scar carcinoma group, the expression (optical density and positive area) of Cx32mRNA were statistically significant (P<0.01). But No statistic differences of their expression were measured between normal skin group and pathological skin scar group. (4) The mutation of Cx26gene was analyzed by direct sequencing,7 target DNA fragments of 28 samples were amplified with which contains six skin scars and one carcinoma tissue, and detected two polymorphism sites in them by DNA sequencing. (5) The mutation of Cx32gene was analyzed by direct sequencing,6 target DNA fragments of 28 samples were amplified with which contains four skin scars and two carcinomas and detected none mutation in them by DNA sequencing. Conclusions:(1) The low expression of Cx26 and its mRNA, Cx32 and its mRNA may be play important roles in the process of the occurrence and development in skin scar carcinoma tissues. (2) There is higher expression of Cx26 and its mRNA in the skin scar than normal epitheliums, may be its related to the hyperplasia of pathological skin scar epitheliums and induce ulceration after skin damaged. (3) Skin scar carcinoma may be diagnosed in the earlier period by detecting the decreased level expression of Cx32mRNA in the scar epithelial,so it is worth more attention. (4) With find out any pathogenic gene mutation sites in skin scar and scar carcinoma tissues of Cx26 and Cx32,maybe they play little important role in carcinogenesis of scar carcinomas.
