A quantum dot is a semiconductor nanocrystals . Compared with traditional organic fluorescent dyes , it has the emission spectra narrow and symmetrical , wide excitation spectra and fluorescence lifetime long , good dispersion with stability and has a significant resistance to photobleaching outstanding fluorescence properties . Detected in the water phase of the quantum dots directly synthesized and applied to bacteria as the goal, mainly in the following two aspects : 1 . Direct synthesis of water-soluble quantum dots . Processed in the organic phase synthesis of quantum dots , the harsh reaction conditions , such as non-water soluble , the present study NaHSe as a precursor , thioglycolic acid as stabilizer as well as anaerobic, and stirred under an atmosphere of direct synthesis of the water - soluble quantum dot . PH, stabilizer , and its amount of fluorescent quantum dots , quantum dot fluorescence spectral lines , transmission electron microscopy were used to characterize . Experimental results show that the method can be rapid synthesis under simple conditions , good stability, good fluorescence properties of quantum dots . 2 . Detection of Bifidobacterium species-specific 16S rDNA. Lee constructed a detection of Bifidobacterium species-specific the 16S rDNA new method . Carboxy - modified 525nm quantum dot (QD) with an amino - modified DNA in effect , the EDC formed QDDNA complexes connected by dehydration and as a fluorescence resonance energy transfer system of the donor , the organic fluorescent groups ROX modified DNA as a fluorescence resonance energy transfer system receptors and Bifidobacterium species - specific 16S rDNA hybridization detection probe . When the probe hybridize with the target sequence is shortened as the distance between the donor 525nm QD as a receptor of ROX to the effective distance of the fluorescence resonance energy transfer occurs . In this case , to not cause ROX emitting a wavelength excitation light emission of the quantum dots , the fluorescence intensity decreased , while the increase in fluorescence intensity of the ROX . Such changes in fluorescence intensity in the case of the target sequence does not exist , does not occur . QD and ROX fluorescence intensity , change is this detection system is quick , simple guarantee .
