The spruce bark beetle is a hazard on the Eurasian continent serious forest pest, the pest in China are mainly distributed in Heilongjiang, Jilin, Qinghai. In recent years, some areas in eastern Jilin and Heilongjiang eastern outbreak of the disaster, caused significant damage to the local forestry production and ecological environment. In the Northeast region, China scholars in the past only the insects through the study of biology and ecology, and the results of different scholars are not entirely consistent, combined with China not as important pests of quarantine pest, and therefore has reason to suspect Northeast China spruce bark beetle populations may be mixed with other regions or geographic population differentiation. To explore whether the other regions of the spruce bark beetle populations in northeast China or pest whether differentiation between regional populations, the test mtDNA-12S rRNA gene, mtDNA-CO Ⅰ gene and mtDNA-CO Ⅱ gene on parts of Jilin and Heilongjiang spruce bark beetle species and among populations of Molecular phylogenetic analysis. The tests to establish the optimal reaction conditions for amplification of the insect mtDNA sequences measured had the pest mtDNA-12S rRNA of mtDNA-CO Ⅰ and mtDNA-CO Ⅱ gene sequences, the nucleotide sequence of these three groups of genes transition / transversion conversion ratio of genetic distance variation sites. Neighbor-joining method (NJ) based on these three groups of gene sequences, minimum evolution (ME), maximum parsimony (MP) and the ex pairing (UPGMAM) construct molecular phylogenetic tree of the populations of the different regions and explore the different regions populations kinship. The results show that: the trial did not find outside the spruce bark beetle population, and the area of ??the spruce bark beetle has not generated significant differentiation of populations asked. Northeast China, the unity of the biology of the pest, and laid the foundation for the prevention and control of ecological studies. The article also combined Genbank on other geographic populations of the pest sequence analysis of the sequencing of the experimental results prove that the results of the experiment are not nuclear mitochondrial pseudogenes. The article also combined Genbank on pest all mtDNA-CO Ⅰ gene sequences constructed molecular phylogenetic trees. The geographical location of the phylogenetic tree and insect created basically consistent. The achievements of the results showed that the insect has two geographic populations: European stocks and Asian populations, where the Asian population is constituted by the population of our country and the population of Russia.
