ObjectivesTea-polyphenols extracted from green tea, ester-type catechins seperated from the tea-polyphenols, which (EGCG and ECG) was used as the main drug, meanwhile, making use of this drug to prepare vaginal carrier suppository with ester-catechins to kill the human papillomavirus (HPV), which was able to furnish the experimental foundation for the development of vaginal carrier suppository with ester-catechins to prevent and treat uterine cervix cancer.MethodsThrough optimuming selection for extracting time, extracting temperature, extracting times and volume ratio of water and ethanol, the last, fixing the best extracting condition to get refined tea-polyphenols and silicagel column by using separately chloroform、ethyl acetate and silicone, of which content was determented with RP-HPLC. Using glycerinated gelatin、cetomacrogol 400、stearic acid hydrocarbon polymer oxygen (40) ester and fatty acid mixture as substrate, through observing and studying appearance、in vitro dissolution test as well as stripping accumulation percentage, screening out the suitable substrate to prepare vaginal carrier suppository with ester-catechins.ResultsBy means of single-factor test and orthogonal test, the best extraction condition of tea-polyphenols was that time、temperature、times and volume of water and ethanol were respectively 60min、70℃、3 and 2:1, The content of EGCG was 70.5%,ECG14.1%,extraction ratio was 8.46% and 1.69%, finally, fatty acid mixture becomed the best substrate. ConclusionOn condition that volume of water and ethanol (2:1) as extraction solvent, extraction time(60min)、temperature(70℃)and extraction times(3). The higher levels of ester-catechins was got by separating tea-polyphenols, furthermore ester-catechins came up to Pharmacopoeia of People’s Republic of China(CHP).
