Synergistic Effect of TRAIL and Adriamycin on Apoptosis of Osteosarcoma Cell Line MG-63 in Vitro

Objective:Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is a member of tumor necrosis factor (TNF) family. TRAIL could specifically induce apoptosis in various tumor cells, and is nontoxic to normal tissues and cells. Because not all tumor cells are sensitive to TRAIL, more and more researches are focusing on the method of combination application of TRAIL and radiation or chemotherapeutic agents recently. One aim of this study is to investigate the effect of TRAIL inducing apoptosis on osteosarcoma cell line MG-63 in vitro. Another aim is to determine if there is a synergistic effect of combined TRAIL with adriamycin (ADM) and to find out the possible mechanism, in order to provide a new strategy in treating osteosarcoma.Method:The osteosarcoma cell line MG-63 were cultured in the regular condi- tion. Respectively inhibition rate and apoptosis rate were analyzed by MTT and Flow Cytometry (FCM). The morphologic changes and characters of apoptosis were observed by inverted phase contrast microscope. The mRNA expression of cell-surface receptors DR4, DR5, caspase-8, Bcl-2 were examined by RT-PCR. Statistic analysis was processed by SPSS13.0 software.Result:The result of MTT and FCM showed that both TRAIL and ADM can inhibit MG-63 cells. There was a synergistic effect of combined TRAIL with ADM. Under inverted phase contrast microscope,some cells became to be small and round with intact cellular membrane 24 hours after treated with TRAIL. Cellular fragments can be seen when treated with TRAIL combing ADM. The result of RT-PCR showed that, compared with TRAIL alone group,ADM could up-regulate the level of DR4 DR5 and caspase-8 mRNA expression in MG-63 cells in combination group(P<0.05). The mRNA expression of Bcl-2 was no significant change(P>0.05).Conclusion:1.TRAIL could inhibit MG-63 cells by inducing apoptosis in the dose dependent manner.2.TRAIL combining with ADM had a synergistic effect in inducing apoptosis in MG-63 cells.3 .The cause mechanism of synergistic effect may because of the up-regulating of the level of DR4 DR5 and caspase-8 mRNA expression.
